摘  要:目的了解MDCK细胞(Madin-Darby Canine Kidney cells,狗肾细胞)培养法和鸡胚双腔培养法分离流行性感冒(流感)病毒的情况,为佳木斯市流感防控提供病原学资料。方法采用MDCK细胞培养法和鸡胚双腔培养法对811份核酸阳性样本进行流感病毒分离。结果6个监测年中MDCK细胞培养法分离流感病毒,每个年份之间分离率差异有统计学意义(χ^(2)=64.640,P<0.01);分离率最高的年份为2015.4.1—2016.3.31,分离率为60.00%(87/145)。鸡胚双腔培养法分离流感病毒,各年监测中分离率差异有统计意义(χ^(2)=18.429,P<0.01),2019.4.1—2020.3.31分离率最高,为33.87%(21/62)。MDCK细胞培养法分离率优于鸡胚双腔培养法[40.69%(330/811)比21.30%(95/446)],两种培养流感病毒方法的分离率之间差异有统计学意义(χ^(2)=48.343,P<0.01)。MDCK细胞培养法分离出甲型H1N1、H3N2和乙型BV、BY;鸡胚双腔培养法分离出甲型H1N1和乙型BV、BY。MDCK细胞培养法血凝滴度≥32的样本,鸡胚双腔培养法更容易分离出毒株。两种培养方法都在二次传代分离出毒株数最多。结论MDCK细胞培养法流感病毒分离率优于鸡胚双腔培养法;提高鸡胚双腔培养法分离率,重点关注MDCK细胞培养法血凝滴度≥32的样本,进一步优化两种方法病毒传代的工作。Objective To understand the isolation of influenza virus by MDCK cells(Madin-Darby Canine Kidney cells)culture and chicken embryo double chamber culture,so as to provide pathogenic data for influenza prevention and control in Jiamusi City.Method Influenza viruses were isolated from 811 nucleic acid positive samples by MDCK cell culture method and chicken embryo double chamber culture.Results The isolation rate of influenza virus by MDCK cell culture was significantly different among different years(χ^(2)=64.640,P<0.01);the highest isolation rate was 60.00%(87/145)from April 1,2015 to March 31,2016.The isolation rate of influenza virus by chicken embryo double chamber culture was statistically significant among different years(χ^(2)=18.429,P<0.01);the highest isolation rate was 33.87%(21/62)from April 1,2019 to March 31,2020.The isolation rate of MDCK cell culture method was higher than that of chicken embryo double chamber culture method[40.69%(330/811)vs.21.30%(95/446)],with a statistically significant difference between the two methods(χ^(2)=48.343,P<0.01).Type A H1N1,H3N2 and type B BV and BY were isolated by MDCK cell culture;type A H1N1 and type B BV and BY were isolated by chicken embryo double chamber culture.When the hemagglutination titer of MDCK cell culture method was more than or equal to 32,the virus strain was easier to be isolated by chicken embryo double chamber culture method.The numbers of virus plants isolated by the two culture methods were the largest in the second passage.Conclusions The isolation rate of influenza virus by MDCK cell culture method is better than that by chicken embryo double chamber culture method;we should improve the isolation rate of chicken embryo double chamber culture method,focus on the samples with hemagglutination titer more than or equal to 32 in MDCK cell culture method,and further optimize the virus passage of the two methods.
