卡巴胆碱对脓毒症大鼠凝血功能的影响

摘　　要：目的观察卡巴胆碱对脓毒症大鼠凝血功能的影响。方法武汉大学动物实验中心提供的96只SD(Sprague Dawley)大鼠(无特定病原体, 健康雄性)随机分成三组, 每组32只。SHAM组:生理氯化钠溶液(5 ml/kg)左侧股静脉注射;脂多糖(LPS)制备脓毒症模型为LPS组:LPS 10 mg/kg左侧股静脉注射;CBL组:LPS 10 mg/kg+卡巴胆碱10 μg/kg。于各组模型制备前(0 h)及制备后2、4、6 h时间点取8只大鼠, 采血检测肿瘤坏死因子-α(TNF-α)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(Fbg)、D-二聚体。结果注射LPS后TNF-α水平上调, CBL组TNF-α水平低于LPS组(P<0.05)。LPS组在注射LPS后各时间点APTT较SHAM组、CBL组延长(P<0.05), CBL组在6 h时PT较LPS组缩短(P<0.05)。4 h时LPS组较SHAM组、CBL组TT延长(P<0.05), Fbg降低(P<0.05);注射LPS后2、4 h, LPS组较SHAM组、CBL组D-二聚体增加(P<0.05)。结论卡巴胆碱可减轻脓毒症大鼠炎症反应, 改善凝血功能紊乱状态。Objective To investigate the effects of carbachol on coagulation in septic rats.Methods A total of 96 SD(Sprague Dawley)rats(specific pathogen free,healthy male)provided by the Animal Experiment Center of Wuhan University were randomly divided into 3 groups,each with 32 rats.SHAM group:normal saline(5 ml/kg)was injected into the left femoral vein;lipopolysaccharide(LPS)was used to prepare the sepsis model as LPS group:LPS 10 mg/kg was injected into the left femoral vein;CBL group:LPS 10 mg/kg+carbachol 10μg/kg.Eight rats were taken before(0 hour)and 2 hours,4 hours,and 6 hours after the preparation of the models in each group,and blood sampling for detection of tumor necrosis factor-α(TNF-α),prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT)and fibrinogen(Fbg),D-dimer.Results The level of TNF-αwas increased after LPS injection,and the level of TNF-αin the CBL group was lower than that in the LPS group(P<0.05).The APTT in the LPS group was longer than that in the SHAM group and the CBL group at each time point after LPS injection(P<0.05),and the PT in the CBL group was shorter than that in the LPS group at 6 hours(P<0.05).At 4 hours,compared with SHAM and CBL groups,TT was longer in LPS group(P<0.05),and Fbg was lower(P<0.05);at 2 and 4 hours after LPS injection,D-dimer increased in LPS group compared with SHAM group and CBL group(P<0.05).Conclusions Carbachol can alleviate the inflammatory response and improve the coagulation disorder in septic rats.
