摘  要:目的:观察冰片对大鼠离体胸主动脉的舒张作用并探讨其作用机制。方法:采用离体血管环试验方法,经生物信号采集与分析系统测定血管环张力变化,以累积给药法加入冰片,使其终浓度依次为0.3、0.6、0.9、1.2、1.5、1.8μmol/L,研究冰片对去甲肾上腺素(NA)预收缩胸主动脉环张力的作用;通过各种工具药干预,探讨冰片舒张血管作用与内皮、钾通道及钙通道的相关机制。结果:冰片对基础状态下血管张力无明显影响(P>0.05);对NA预收缩的血管有浓度依赖性的舒张作用(P<0.01),且对去内皮血管的舒张作用明显强于内皮完整血管(P<0.05);经亚甲基蓝(MB)、吲哚美辛(INDO)处理后,与模型对照组比较,冰片的舒血管作用均无明显差异(P>0.05);经4-氨基吡啶(4-AP)、5-羟基癸酸(5-HD)、氯化钡处理后,与模型对照组比较,冰片的舒血管作用亦均无明显差异(P>0.05);冰片对高浓度(60 mmol/L)氯化钾引起的血管环收缩具有显著的抑制作用(P<0.01);在无钙环境下,用NA预收缩血管环后,随着加入的Ca;浓度逐渐增加,血管收缩率呈浓度依赖性增加,而用冰片孵育20 min后可使CaCl_(2)缩血管的量效曲线向下移位,最大收缩率降低;在无钾环境下,冰片对NA引起的血管环收缩有显著抑制作用(P<0.01)。结论:冰片对大鼠离体胸主动脉有浓度依赖性的舒张作用,其机制可能与血管平滑肌细胞的电压依赖性钙通道(VDCC)、受体操纵性钙通道(ROCC)及其介导的外钙内流和内钙释放有关。而与内皮细胞一氧化氮(NO)/环磷酸鸟苷(cGMP)途径、前列环素(PGI_(2))途径、电压敏感型钾通道(K_(V))、ATP敏感型钾通道(K_(ATP))以及内向整流型钾通道(K_(IR))无关。Objective:To investigate the vasodilation effect and mechanism of borneol on the isolated thoracic aorta of rats.Methods:Isolated aortic ring assay was used to determine changes of vascular ring tension with a multichannel acquisition and analysis system.Borneol was added cumulatively with final concentrations of 0.3,0.6,0.9,1.2,1.5,and 1.8μmol/L to observe the effect of borneol on aortic rings pre-contracted with noradrenaline(NA).The underlying mechanism of vasodilation effect of borneol on the endothelium,K;channel and Ca;channel was discussed by means of intervention with drugs.Results:Borneol showed no significant effect on the basal tension in rat aortic rings(P>0.05).Borneol could relax NA pre-contracted rings in a concentration-dependent manner(P<0.01),with the effect on endothelium-denuded aorta stronger than that on endothelium-intact aorta(P<0.05).After pre treatment with indomethacin(INDO)and methylene blue(MB),compared with the model group,no significant vasodilation effect was observed in the borneol groups(P>0.05).After incubation with 4-aminopyridine(4-AP),5-hydroxy decanoic acid(5-HD),and BaCl_(2),compared with the model group,no significant difference was also observed in the borneol groups(P>0.05).Borneol significantly inhibited the constriction of aortic rings induced by potassium chloride(60 mmol/L)(P<0.01).In the Ca;-free solution under NA pre incubation,as the concentration of CaCl_(2)gradually increased,the vasoconstriction rate increased in a concentration dependent manner.After pretreatment with borneol for 20 min,the dose-response curve of CaCl_(2)induced vasoconstriction was down shifted,while the maximum contraction rate decreased.In the K;-free solution,borneol significantly inhibited NA induced aortic ring contraction(P<0.01).Conclusion:Borneol can relax NA pre-contracted rings in a concentration-dependent manner,which may be associated with the inhibition of voltage-dependent calcium channels(VDCC),receptor operated calcium channels(ROCC),calcium influx,and calcium release,but it
