摘  要:目的:分析不同剂量黄芩提取物对牙周膜干细胞增殖、凋亡、周期的影响。方法:分离培养牙周膜干细胞,制备黄芩提取液。将细胞分为空白对照组、黄芩提取物0.1、1、10μg/mL组,空白对照组使用细胞培养液做常规细胞培养,黄芩提取物各浓度组细胞培养液中分别加入含有0.1、1、10μg/mL的黄芩提取液。分析黄芩提取物对牙周膜干细胞增殖、凋亡、周期的影响及作用机制。结果:与空白对照组相比,黄芩提取物0.1、1、10μg/mL组细胞增殖率升高,细胞凋亡率降低,G0/G1期细胞减少,S期、G2/M期细胞增多,JNK、p-JNK、Ki67、PCNA、Cyclin D1蛋白表达显著上调(P<0.01),其中以10μg/mL最为显著。结论:黄芩提取物可促进牙周膜干细胞增殖,抑制细胞凋亡,其作用机制可能与激活JNK信号通路相关,且呈现为剂量依赖特征。Objective:To analyze the effects of Scutellaria baicalensis extract(SBE)at different doses on the proliferation,apoptosis,and the cycle of periodontal ligament stem cells(PDLSCs).Methods:The PDLSCs were isolated and cultured and SBE was prepared.The cells were divided into a blank control group and SBE(0.1,1,and 10μg/mL)groups.Routine cell culture was adopted in the blank control group with culture medium,while the cells in the SBE groups were cultured in the medium containing 0.1,1,and 10μg/mL SBE.The effects of SBE at different doses on proliferation,apoptosis,and cycle of PDLSCs were analyzed,and the underlying mechanism was explored.Results:Compared with the blank control group,the SBE groups showed increased cell proliferation,decreased apoptosis,reduced cells in the G0/G1 phase,increased cells in the S phase and the G2/M phase,and up-regulated protein expression of JNK,p-JNK,Ki67,PCNA,and Cyclin D1(P<0.01).The effects were predominant in the high-dose SBE group.Conclusion:SBE can promote the proliferation and inhibit apoptosis of PDLSCs,and the underlying mechanism may be related to the activation of the JNK signaling pathway in a dose-dependent manner.
