摘  要:目的探讨半枝莲药材中黄酮类成分的提取及含量的测定。方法采用超声提取方式对半枝莲中黄酮类成分进行检测,高效液相色谱法对提取的黄酮类成分中的野黄芩苷、木犀草素进行含量测定。结果①本实验超声提取的条件:超声提取时间为30 min,温度为65℃,乙醇浓度为45%,黄酮类提取物的测得率为13.12%;②高效液相色谱条件为:FortisC_(18)色谱柱(4.0 mm×150 mm,2.5μm),流动相为A:甲醇,B:乙腈,C:0.2%磷酸水缓冲溶液梯度洗脱,柱温为28℃,检测波长225 nm,流速为1.0 mL/min,进样量为15μL;野黄芩苷线性关系方程:Y=2.8165×10^(4)X+0.0632×10^(2)(r=0.9997),木犀草素线性关系方程:Y=3.2165×10^(4)X-0.0217×10^(2)(r=0.9994),野黄芩苷在9.87~250.21 mg/L,木犀草素在1.14~29.32 mg/L范围内线性关系良好。结论超声提取在时间为30 min,温度为65℃,乙醇浓度为45%时半枝莲中黄酮类成分的提取率较好,对含量的测定采用高效液相色谱法进行,方法简单,易行,重复性好,稳定性高,是含量测定的最佳方式。Objective To investigate the extraction and determination of flavonoids in Banzhilian(Herba Scutellariae Barbatae).Methods Ultrasonic extraction was used to detect flavonoids in Banzhilian(Herba Scutellariae Barbatae).HPLC was used to determine the contents of lutein and luteolin in the extracted flavonoids.Results①Conditions for ultrasonic extraction in this experiment:ultrasonic extraction time was 30 min,temperature was 65℃,ethanol concentration was 45%,and the measured rate of flavonoid extract was 13.12%.②The conditions of HPLC:Fortis C_(18) column(4.0 mm×150 mm,2.5μm)was used.The mobile phase was gradient elution of A(methanol),B(acetonitrile)and C(0.2%aqueous phosphate buffer solution).The column temperature was 28℃.The detection wavelength was 225 nm.The flow rate was 1.0 mL/min and the injection volume was 15μL.The linear relationship equation of glucoside was Y=2.8165×10^(4)X+0.0632×10^(2)(r=0.9997).The linear relationship equation of luteolin was Y=3.2165×10^(4)X-0.0217×10^(2)(r=0.9994).Baicalin had a good linear relationship in the range of 9.87~250.21 mg/L and luteolin had a good linear relationship in the range of 1.14~29.32 mg/L.Conclusion The ultrasonic extraction time is 30 min,the temperature is 65℃and the ethanol concentration is 45%.The extraction rate of flavonoids in Banzhilian(Herba Scutellariae Barbatae)is better.The content is determined by high performance liquid chromatography.The method is simple,easy and of good repeatability with high stability.It is the best way to determine the content.
