摘 要:目的研究雷公藤多苷对子宫内膜异位症(EMs)在位内膜间质细胞(ESC)增殖和凋亡的影响,并探讨其分子机制。方法运用细胞计数试剂盒(CCK-8)和集落形成实验检测雷公藤多苷对EMs ESC细胞增殖的影响。流式细胞术检测细胞凋亡。实时荧光定量PCR检测KISS-1 mRNA表达量。蛋白质印记(Western Blot)检测Ki67、半胱氨酸蛋白酶3前体(Pro-Caspase3)、裂解的Caspase3(Cleaved-Caspase3)和B细胞淋巴瘤相关x蛋白(Bax)表达量。转染KISS-1小干扰RNA至EMs ESC细胞,检测干扰KISS-1表达对雷公藤多苷处理的EMs ESC细胞增殖和凋亡的影响。结果雷公藤多苷处理后EMs ESC细胞活力、克隆形成数、Ki67和Pro-Caspase3蛋白表达显著降低,凋亡率、Cleaved-Caspase3和Bax蛋白表达显著升高,KISS-1 mRNA表达显著升高(P<0.05)。干扰KISS-1表达后,雷公藤多苷处理的EMs ESC细胞活力、克隆形成数、Ki67和Pro-Caspase3蛋白表达显著升高,凋亡率、Cleaved-Caspase3和Bax蛋白表达显著降低(P<0.05)。结论雷公藤多苷可抑制EMs ESC细胞增殖,并促进其凋亡,其机制可能与上调KISS-1表达有关。Objective To study the effect of Tripterygium polyglycosides on the proliferation and apoptosis of endometrial stromal cells(ESCs)in endometriosis(EMs),and further explore its molecular mechanism.Methods The cell counting kit(CCK-8)and colony formation experiment were selected to detect the effects of Tripterygium glycosides on the proliferation of EMs ESC cells.Flow cytometry detected the apoptosis.Real-time quantitative PCR tested the expression level of KISS-1 mRNA.Western blot measured the expression of Ki67,Pro-Caspase3,Cleaved-Caspase3 and B cell lymphoma-associated x protein(Bax).KISS-1 small interfering RNA was transfected into EMs ESC cells,and the effects of interfering with KISS-1 on the proliferation and apoptosis of EMs ESC cells treated with Tripterygium glycosides were examined by the above methods.Results After treated with Tripterygium glycosides,EMs ESC cell viability,clone formation number,expression of Ki67 and Pro-Caspase3 proteins were significantly reduced,the apoptosis rate and expressions of Cleared-Caspase3 and Bax proteins were significantly increased,and KISS-1 mRNA expression was significantly increased(P<0.05).After interfering with the expression of KISS-1,the cell viability,clone formation number,expression of Ki67 and Pro-Caspase3 proteinsin EMs ESC cells treated with Tripterygium glycosides were significantly increased,while the apoptosis rate and the expressions of Cleaved-Caspase3 and Bax proteins were significantly reduced(P<0.05).Conclusion Tripterygium glycosides can inhibit EMs ESC cell proliferation and promote its apoptosis,the mechanism may be related to the up-regulation of KISS-1 expression.
