摘 要:目的研究白细胞介素(IL)-6体外对SZ95人皮脂腺细胞脂质合成的影响。方法CCK8法及尼罗河红染色法检测IL-6对细胞增殖和细胞脂质合成的影响,实时荧光聚合酶链反应(PCR)及蛋白印迹法检测IL-6对脂质合成相关基因(PPARG、SREBP1和FAS)表达的影响。结果10 ng/ml IL-6作用24 h和48 h后细胞活力百分率分别为120%和126%,10 ng/ml及1 ng/ml IL-6作用48 h后细胞中性脂质百分率分别为125%和116%,均显著高于对照组(P<0.05);伴随PPARG、SREBP1和FAS mRNA及蛋白表达上调。结论IL-6体外促进SZ95人皮脂腺细胞脂质合成,显示IL-6可能通过促进皮脂腺细胞脂质合成而参与寻常痤疮等相关疾病的发生。Objective To evaluate the effect of IL-6 on lipogenesis of human SZ95 sebocytes in vitro. Methods CCK8 and Nile red staining methods were performed to detect cell viability and lipid contents. Real-time fluorescence-based PCR and Western blot were used to detect lipogenesis-related genes expression. Results After treated with 10 ng/ml IL-6 for 24 h and 48 h, cell viability percentage were 120% and 126%, respectively;after incubated with 10 and 1 ng/ml IL-6 for 48 h, cell neutral lipids percentages were 125% and 116%,respectively, which were significantly higher than controls(P<0.05);the above process accompanied by the up-regulated expression of PPARG, SREBP1 and FAS. Conclusion IL-6 may promote lipogenesis of SZ95 human sebocytes in vitro, indicating that IL-6 may participate in the occurrence of acne vulgaris and other related diseases by promoting lipogenesis of human sebocytes.
