摘  要:目的观察白芍总苷(total glueosides of paeony, TGP)对缺血后再灌注乳鼠心肌细胞细胞焦亡的影响。方法乳鼠心肌细胞随机分为正常对照组、模型组及低、中、高剂量TGP组(TGP终浓度分别为25,50,100 mg/L),测定收集细胞上清液中LDH含量;以MTT法检测每组活细胞数目;流式细胞仪检测各组心肌细胞焦亡率;Western Blotting检测心肌细胞焦亡相关斑点样蛋白(ASC)、NLRP3、半胱氨酸天冬氨酸特异蛋白酶1(caspase-1)以及白细胞介素1β(IL-1β)表达。结果与缺血再灌注组相比,随白芍总苷浓度提升,心肌细胞缺血再灌注后的细胞活力升高,而LDH分泌量,焦亡率,NLRP3、ASC、caspase-1、IL-1β的表达均下降,结论白芍总苷对大鼠心肌缺血再灌注损伤的保护作用明显,可通过caspase-1途径来抑制缺血再灌注心肌细胞的焦亡。Objective To investigate the effects of total glucosides of paeony on rat cardiac myocyte pyroptosis induced by ischemia-reperfusion. Methods SD neonatal rats were randomly divided into 5 groups: control group, I/R group, TGP low-dose group(25 mg/L), TGP middle-dose group(50 mg/L), and TGP high-dose group(100 mg/L). Cultured supernatants were collected to measure lactic dehydrogenase(LDH).The viability of cardiomyocytes was assayed by MTT. The pyroptosis rate of cardiomyocytes was determined by flow cytometry. The expressions of NLRP3, ASC, caspase-1, and IL-1β were measured by Western Blot. Results Compared with the I/R group, with the increase of total glucosides of paeony, the cell viability of cardiomyocytes after ischemia-reperfusion increased in the TGP group, while the secretion of LDH, the pyroptosis rate, the expression of NLRP3, ASC, caspase-1, and IL-1β were lower. Conclusion TGP has an obvious protective effect on myocardial ischemia-reperfusion injury in rats, which may be inhibited myocardial pyroptosis through caspase-1 pathway.
