摘  要:目的研究二至丸的HPLC指纹图谱及其提取物的体外抗氧化活性。方法采用Aglient Zorbax SB-C;色谱柱(250 mm×4.6 mm, 5μm),流动相为乙腈-0.2%磷酸水,梯度洗脱,温度30℃,流速1 mL·min^(-1),检测波长224 nm;用抗坏血酸作阳性对照,采用Frap、ABTS、DPPH法分别测定1~1×10;μg·mL^(-1)二至丸醇提冻干粉水溶液的体外抗氧化能力。结果 12批二至丸共指认出11个共有峰,相似度为0.795~0.984;聚类分析将12批二至丸分成3类,不同产地与批次的药材所制备的二至丸具有明显差异;主成分分析筛选出2个主成分,累计贡献率达99.92%,表明前两个主成分能够反应二至丸的基本特征和大部分信息。抗氧化作用结果显示:二至丸的Frap为0.116 mmol·mL^(-1);DPPH自由基清除率可达79.06%,IC;为0.077±0.005 mg·mL^(-1);ABTS清除率可达59.40%,IC;为0.707±0.05 mg·mL^(-1)。结论所用方法简单可靠,体外抗氧化实验证明二至丸醇提物有一定的抗氧化作用。OBJECTIVE To study the fingerprint and antioxidant activity in vitro of Erzhi pills.METHODS Aglient Zorbax SB-C;(250 mm×4.6 mm, 5 μm) column was used for fingerprint analysis.The mobile phase composed of acetonitrile-0.2% phosphate acid solution in gradient elution manner with a flow rate of 1.0 mL·min^(-1),temperature of 30 ℃ and the detection wavelength of 224 nm.Erzhi pills in range of 1-1×10;μg·mL^(-1)were used to extract alcohol freeze-dried powder aqueous solution as the sample, and ascorbic acid was used as the positive control.Frap, ABTS and DPPH methods were used to determine the antioxidant capacity of Erzhi pills in vitro.RESULTS A total of 11 common peaks were identified in 12 batches of Erzhi pills, and the similarity of fingerprints was 0.795-0.984.In cluster analysis, 12 batches of Erzhi pills were divided into 3 categories, and the results showed that there were significant differences in Erzhi pills prepared from different origins and batches of medicinal materials.Principal component analysis screened out 2 principal components.It showed that the first two principal components could reflect the basic characteristics and most information of Erzhi pills.The cumulative contribution rate of the first two components reached 99.92%.The results of antioxidant activity showed that Frap of Erzhi pills was 0.116 mmol·L;.DPPH radical scavenging rate was up to 79.06% and IC;was 76.608±4.968 μg·mL^(-1).ABTS clearance rate was 59.40% and IC;was 0.707±0.05 mg·mL^(-1).CONCLUSION This method is simple and reliable, and the antioxidant experiment in vitro proves that the alcohol extract of Erzhi pills has certain antioxidant effect.
