摘 要:目的利用小鼠盲肠结扎穿刺模型探讨脓毒症晚期髓系造血情况。方法将C57 BL/6小鼠分成假手术组和脓毒症组,采用盲肠结扎穿刺术(CLP)建立小鼠脓毒症模型。利用流式细胞仪检测CLP术后12d脓毒症小鼠骨髓和脾脏造血前体细胞比例的变化情况。结果与假手术组相比,CLP术后12d小鼠骨髓中的GMPs(粒-单核细胞祖细胞)、MPPs(多潜能祖细胞)、LSKs(造血干细胞Lin^(-)Sca-1^(+)c-Kit^(+))的比例显著增加;脾脏中GMPs、LSKs的比例也明显增加。结论脓毒症晚期小鼠髓系造血显著增加。Objective To investigate effects of sepsis during the late stage on myelopoiesis with mouse model which received cecal ligation and puncture(CLP).Methods The studied mice were divided into two groups.The sepsis model was developed by CLP,while Sham operation was conducted in the control group.Changes of hematopoietic stem cells and precursor cells in bone marrow and spleen were analyzed with flow cytometer 12 days after CLP.Results Compared with control group,the proportion of GMPs(granulocyte/monocyte progenitors),MPPs(multipotent progenitors),and LSKs(hematopoietic stem cells,Lin^(-)Sca-1^(+)c-Kit^(+))in bone marrow of the sepsis group increased.Meanwhile,sepsis mice also showed an increased frequency of GMPs and LSK in spleen.Conclusion Myelopoiesis of sepsis mice druing the late stage increases dramatically in both bone marrow and spleen.
