摘 要:目的探讨祛风地黄汤对银屑病模型小鼠皮损的改善作用。方法将60只BALB/c小鼠随机分为空白对照组(等体积0.9%氯化钠溶液)、模型组(等体积0.9%氯化钠溶液)、甲氨蝶呤组(1mg/kg),以及祛风地黄汤低、中、高剂量组(8.76,17.55,35.10g/kg),各10只。通过局部涂抹咪喹莫特以复制银屑病小鼠模型。建模成功后,各组小鼠灌胃相应药物或等体积0.9%氯化钠溶液。以银屑病皮损面积及严重程度指数(PASI)评估小鼠皮损情况;以苏木素-伊红(HE)染色观察小鼠皮肤组织病理形态学;以酶联免疫吸附法检测小鼠血清中肿瘤坏死因子-β(TNF-β)、白细胞介素10(IL-10)、白细胞介素17(IL-17)、白细胞介素6(IL-6)水平;以免疫组织化学法检测小鼠皮损组织中增殖细胞核抗原(PCNA)及CD_(3)^(+)的表达水平;以Westernblot法检测小鼠皮损组织中叉头样转录因子3(Foxp3)及孤独核受体(RORγt)蛋白的表达水平。结果与模型组比较,甲氨蝶呤组和祛风地黄汤低、中、高剂量组小鼠皮损组织表皮层厚度均显著减小,真皮层炎性细胞浸润程度均显著减轻;小鼠PASI评分和IL-17,IL-6水平,皮损组织PCNA,CD_(3)^(+)表达水平及RORγt蛋白表达水平均显著降低;IL-10、TNF-β及Foxp3蛋白表达水平均显著升高(P<0.05)。结论祛风地黄汤对模型小鼠银屑病样皮损的改善作用明显,其机制可能与辅助性T细胞17/调节性T细胞失衡有关。Objective To investigate the effect of Qufeng Dihuang Decoction on skin lesions in psoriasis model mice.Methods A total of 60 BALB/c mice were randomly divided into blank control group(equal volume of 0.9%Sodium Chloride Solution),model group(equal volume of 0.9%Sodium Chloride Solution),methotrexate group(1 mg/kg)and Qufeng Dihuang Decoction low-,medium-and high-dose groups(8.76,17.55,35.10 g/kg),10 rats in each group.Local application of imiquimod was used to replicate the psoriasis mice model.After successful modeling,mice in each group were gavaged with corresponding drugs or equal volume of 0.9%Sodium Chloride Solution.The skin lesions of mice were evaluated by the Psoriasis Area and Severity Index(PASI),the pathomorphology of skin tissue was observed by the hematoxylin-eosin(HE)staining,the levels of serum tumor necrosis factor-β(TNF-β),interleukin-10(IL-10),interleukin-17(IL-17)and interleukin-6(IL-6)were detected by the enzyme-linked immunosorbent assay(ELISA),the expression levels of proliferating cell nuclear antigen(PCNA)and CD_(3)^(+) in the tissue of skin lesions were detected by immunohistochemistry,the expression levels of forkhead like transcription factor 3(Foxp3)and retinoic receptor-related orphan receptor gamma-t(RORγt)protein in the tissue of skin lesions were detected by Western blot.Results Compared with those in the model group,the thickness of epidermal layer and the degree of inflammatory cell infiltration of dermis were significantly reduced in methotrexate group and Qufeng Dihuang Decoction low-,medium-and high-dose groups,the PASI score and the levels of IL-17 and IL-6 decreased significantly,the expression level of PCNA,CD_(3)^(+) and RORγt protein in skin lesions decreased significantly,and the levels of IL-10 and TNF-β,the expression level of Foxp3 protein increased significantly(P<0.05).Conclusion Qufeng Dihuang Decoction can significantly improve psoriasis-like lesions in model mice,and its mechanism may be related to the imbalance of Th17/Treg cells.
