摘 要:目的探究人参皂苷Rg1对冠心病大鼠心肌细胞凋亡的影响并分析其作用机制。方法选无特定病原体(SPF)级健康雄性SD大鼠50只,冠心病造模成功后,随机分为对照组、模型组、人参皂苷Rg1低剂量组、人参皂苷Rg1中剂量组以及人参皂苷Rg1高剂量组,每组10只。对照组常规饮食,不进行冠心病造模,给予人参皂苷Rg1等体积生理盐水灌胃,每日1次;模型组构建冠心病模型,给予人参皂苷Rg1等体积生理盐水灌胃,每日1次;人参皂苷Rg1低剂量组、人参皂苷Rg1中剂量组、人参皂苷Rg1高剂量组均构建冠心病模型,术后分别给予50 mg/kg、75 mg/kg、100 mg/kg人参皂苷Rg1灌胃,每日1次。所有实验动物均连续灌胃12周。光学显微镜下观察细胞形态;酶联免疫吸附测定法(ELISA)检测白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量;试剂盒检测超氧化物歧化酶(SOD)及丙二醛(MDA)含量;逆转录聚合酶链式反应(RT-PCR)和蛋白免疫印迹(Western Blot)法检测Bax、Bcl-2、Caspase-3等凋亡相关蛋白及mRNA表达。结果光镜下模型组大鼠心肌组织细胞形态发生明显改变,细胞体积明显缩小,核固缩明显,人参皂苷Rg1低、中、高剂量组大鼠心肌组织细胞水肿状态均较模型组有所改善,人参皂苷Rg1高剂量组细胞肿胀程度缓解最为明显,细胞形态基本趋于正常。与对照组比较,模型组及人参皂苷Rg1低、中、高剂量组IL-1β、IL-6、TNF-α、MDA水平均明显升高(P<0.05),SOD活性明显降低(P<0.05);与模型组比较,人参皂苷Rg1低、中、高剂量组IL-1β、IL-6、TNF-α、MDA水平明显降低(P<0.05),SOD活性明显升高(P<0.05);与人参皂苷Rg1中、低剂量组比较,人参皂苷Rg1高剂量组IL-1β、IL-6、TNF-α、MDA水平明显降低(P<0.05),SOD活性明显升高(P<0.05)。与对照组比较,模型组及人参皂苷Rg1低、中、高剂量组Bax及Caspase-3蛋白及mRNA表达水平明显升高(P<0.05),Bcl-2蛋Objective To investigate the effect of ginsenoside Rg1 on myocardial apoptosis in rats with coronary heart disease.Methods After the successful modeling of coronary heart disease,fifty healthy male Sprague-Dawley(SD)rats without specific pathogen(SPF)were randomly divided into control group,model group,ginsenoside Rg1 low-dose group,ginsenoside Rg1 medium-dose group and ginsenoside Rg1 high-dose group according to body weight,with 10 rats in each group.The control group was given the same volume of physiological saline with ginsenoside Rg1 by gavage,once a day.Model group was given ginsenoside Rg1 equal volume of normal saline,once a day.Low dose ginsenoside Rg1 group,medium dose ginsenoside Rg1 group,and high dose ginsenoside Rg1 group were given 50 mg/kg,75 mg/kg,and 100 mg/kg ginsenoside Rg1 by gavage,once a day,respectively.All animals were fed intragastrically for 12 weeks.Cell morphology was observed under optical microscope.The contents of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were determined by enzyme-linked immunosorbent assay(ELISA).The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)were detected by the kit.The expression of Bax,Bcl-2,Caspase-3,and other apoptosis-related proteins and genes were detected by reverse transcription polymerase chain reaction(RT-PCR)and Western Blot.Results The myocardial tissue cell morphogenesis of model group rats was changed obviously,cell volume was narrowed significantly with nucleus pycnosis.Compared with model group,the myocardial tissue edema of rats in low,medium,and high-dose ginsenoside Rg1 groups was improved.The cell swelling of ginsenoside Rg1 high-dose group was relieved most obviously,and the cell morphology basically tended to normality.Compared with the control group,the levels of IL-1β,IL-6,TNF-α,and MDA in model group,ginsenoside Rg1 low,medium,and high-dose groups were increased significantly(P<0.05),while SOD activity was decreased significantly(P<0.05).Compared with the model group,the levels o
