摘 要:目的探讨梓醇-川芎嗪(CL)方对阿尔茨海默病(AD)模型小鼠认知功能的影响。方法选用APPswe/PS1dE9双转基因模型小鼠,随机分为模型组、梓醇组、川芎嗪组、CL方组、盐酸多奈哌齐(安理申)组,每组10只。6月龄C57BL/6J小鼠10只作为对照组。梓醇组、川芎嗪组、CL方组及安理申组小鼠每天给予相对应剂量的梓醇、川芎嗪、CL方及安理申灌胃。对照组和模型组小鼠每天给予等体积的生理盐水灌胃,各组均为每天1次,连续灌胃8周,进行行为学测试即Morris水迷宫检测和新物体识别(NOR)检测。结果CL方可以明显缩短APPswe/PS1dE9双转基因小鼠的逃避潜伏期、增加目标象限出现次数、延长目标象限游程、提高目标象限游程比、延长目标象限时间、提高目标象限时间比、增加穿台次数、缩短首次穿越平台时间、提高NOR测试中相对识别指数(P<0.05),以上作用不仅优于梓醇组、川芎嗪组,而且也优于安理申组。结论CL方可以有效改善AD模型小鼠的认知障碍,其效果既优于其单一成分,也优于安理申。Objective To investigate the effect of Catalpol-Ligustrazine(CL)Prescription on the cognitive function of mice model with Alzheimer′s disease(AD).Methods APPswe/PS1dE9 double transgenic mice were randomly divided into model group,Catalpol group(C group),Ligustrazine group(L group),CL group,and donepezil hydrochloride(Aricept)group,with 10 mice in each group.Ten C57BL/6J mice aged 6 months were chosen as control group.Mice in C group,L group,CL group,and Aricept group were given the corresponding doses of Catalpol,Ligustrazine,CL Prescription,and Aricept gavage every day respectively.Mice in the control group and model group were given an equal volume of saline gavage every day,once per day for each group,with continuous gavage for 8 weeks.Then morris water maze(MWM)detection and new object recognition(NOR)detection were performed.Results CL Prescription could significantly shorten the escape latency of APPswe/PS1dE9 double transgenic mice,increase the target quadrant occurrences,extend target run-length,improve target quadrant run ratio,extend the time of target quadrant,improve target quadrant time ratio,increase the crossing table number,shorten the time of first crossing platform,enhance the relative identification index(DI)of NOR test(P<0.05).The effects were superior to those of Catalpol or Ligustrazine or those of Aricept.Conclusion CL Prescription can effectively improve the cognitive impairment of AD model mice,and its effects were not only superior to its single component,but it were superior to Aricept too.
