摘　　要：目的分析青藤碱治疗类风湿性关节炎的免疫抑制作用及机制。方法将青藤碱作用于大鼠模型中,使用MTT法测定脾淋巴细胞增殖反应和滑膜细胞增殖反应、ELISA测定血清抗IL-1β、IL-17和血清TNF-α情况。结果青藤碱在体给药可抑制LPS和PMA诱导的大鼠脾淋巴细胞增殖,IL-1β、IL-17和血清TNF-α水平值比正常的明显升高,对人类风湿性关节炎滑膜细胞增殖也有显著抑制作用。结论青藤碱对于类风湿性关节炎和脾淋巴细胞具有免疫抑制作用,而诱导细胞的凋亡可能为免疫作用机制的一种机制。Objective Analyze the immunosuppressive effect and mechanism of sinomenine in the treatment of rheumatoid arthritis.Methods Sinomenine was applied to the rat model.MTT assay was used to determine the proliferation of spleen lymphocytes and synovial cells,and ELISA was used to determine the anti-IL-1β,IL-17 and serum TNF-αlevels.Results Sinomenine can inhibit the proliferation of rat spleen lymphocytes induced by LPS and PMA in vivo,the levels of IL-1β,IL-17 and serum TNF-αare significantly higher than normal,and it also has significant inhibitory effect on the proliferation of human rheumatoid arthritis synovial cells.Conclusion Sinomenine has immunosuppressive effect on rheumatoid arthritis and splenic lymphocytes,and inducing apoptosis may be one of the mechanisms of immune action.
