摘　　要：目的观察芪玉三龙汤药物血清对人肺癌A549细胞增殖及凋亡的影响。方法制备芪玉三龙汤药物血清,体外培养A549细胞。将A549细胞分为正常血清组、5%药物血清组、10%药物血清组和20%药物血清组。CCK-8法和集落形成实验检测A549细胞增殖,TUNEL法检测细胞凋亡,Western blot检测凋亡相关蛋白Bax、cleaved Caspase-3、Bcl-2表达。结果与正常血清组比较,不同浓度芪玉三龙汤药物血清组A549细胞活力明显降低(P<0.001),细胞凋亡率明显升高(P<0.001),10%药物血清组和20%药物血清组A549细胞Bax、cleaved Caspase-3蛋白表达明显升高,Bcl-2蛋白表达明显降低(P<0.001)。结论芪玉三龙汤可能通过上调A549细胞Bax和cleaved Caspase-3蛋白表达,下调Bcl-2蛋白表达,进而诱导肿瘤细胞凋亡,抑制肿瘤细胞增殖。Objective To explore the effects of Qiyu Sanlong Decoction containing serum on proliferation and apoptosis of human lung cancer A549 cells.Methods Qiyu Sanlong Decoction containing serum was prepared and lung cancer A549 cells were cultured in vitro.A549 cells were divided into normal serum group,5%medicated serum group,10%medicated serum group and 20%medicated serum group.CCK-8 and colony formation assay were performed to detect the proliferation of A549 cells;apoptosis was detected by TUNEL;Western blot was used to detect the expression of apoptosis-related proteins.Results Compared with the normal serum group,different concentrations of Qiyu Sanlong Decoction containing serum inhibited the activity of A549 cells(P<0.001),and the apoptosis rate of A549 cells significantly increased(P<0.001).The proteins expression of Bax and cleaved caspase-3 in 10%medicated serum group and 20%medicated serum group increased significantly,and the expression of Bcl-2 decreased significantly(P<0.001).Conclusion It is possible for Qiyu Sanlong Decoction to increase the proteins expression of Bax and cleaved Caspase-3 in A549 cells,and decrease the expression of Bcl-2,thereby inducing tumor cell apoptosis,and inhibit the proliferation of tumor cells.
