摘 要:探讨钩吻素子(koumine)对LPS诱导的猪肠道上皮细胞(ZYM-SIEC-02)损伤的保护作用及其机制。采用CCK-8法检测钩吻素子对ZYM-SIEC-02细胞增殖的影响,并用LPS建立细胞损伤模型,用CCK-8法检测分析钩吻素子保护ZYM-SIEC-02细胞免受LPS损伤的效果;生化方法检测丙二醛(MDA)含量、总超氧化物歧化酶(TSOD)和过氧化氢酶(CAT)活性;荧光酶标法检测氧自由基(ROS)含量;流式细胞仪检测细胞凋亡率。结果表明,检测浓度范围内钩吻素子对ZYM-SIEC-02细胞无毒性;钩吻素子可以调控ZYM-SIEC-02细胞中TSOD活性、CAT活性、MDA水平和ROS的产生,抑制LPS诱导的ZYM-SIEC-02细胞凋亡,减缓LPS对ZYM-SIEC-02细胞的损伤。提示钩吻素子可保护ZYM-SIEC-02拮抗LPS诱导的损伤,具有一定的剂量依赖关系,其作用机制与抑制细胞凋亡有关。Gelsemium alkaloids have antioxidant effect and are widely used as food additives.Koumine is a monomer of gelsemium alkaloids.In this study,we aimed to evaluate antioxidant capacity of koumine by measuring and analyzing the total superoxide dismutase(T-SOD) activity,catalase(CAT) activity,malondialdehyde(MDA) content and ROS,which are related with antioxidant capacity in LPS-treated ZYM-SIEC-02 cells.Koumine was found to promote the growth of ZYM-SIEC-02 cells measured by the CCK-8 bioassay dose-dependently.LPS could induce ROS and MDA accumulation in ZYM-SIEC-02 cells and concomitant reduce of the enzymatic activities of T-SOD and CAT.The LPS-induced cellular injury could be alleviated by co-treatment with koumine.The rate of apoptosis was measured with flow cytometry,and it was found that LPS could improve the apoptosis rate.The effects could also be alleviated by co-treatment with koumine in ZYM-SIEC-02 cells.The results suggest that koumine may repair LPS-induced injury of ZYM-SIEC-02 cells through counteracting LPS-induced oxidative stress.
