摘  要:设计合成了一种具有肿瘤靶向、可携带si RNA的新型金纳米棒(Gold nanorod,GNR)纳米靶向系统GNR-MAN。GNR长约30 nm,直径约10 nm。NMR表征显示GNR-MAN制备成功,经修饰后的GNR-MAN未见聚集。带正电荷的MUA-PEI代替CTAB对GNR修饰,可载入带负电的si RNA。由于某些肿瘤细胞、免疫细胞等表面甘露糖受体高表达,末端经甘露糖修饰后的金纳米棒可具有靶向这些细胞的功能。A novel nano system was developed,which could target liver cancer and carry siRNA.The first was a GNR backbone that was about 30 nm in length and 10 nm in diameter.No aggregation of GNRs was observed after chemical conjugation.Secondly,GNR was modified by positively charged MUA-PEI instead of CTAB in order to load negatively charged siRNA.The third functional moiety was the mannose(MAN).Considering the abundant expression of MAN receptors on some cancer cells,we conjugated mannose as a guide adaptor to the nano-carrier GNR-MAN,making it capable of tumor-specific homing.
