摘 要:目的评价柴芍六君汤对肝郁脾虚型溃疡性结肠炎(Ulcerative colitis,UC)大鼠炎性反应因子白细胞介素-4(interleukin 4,IL-4)、白细胞介素-10(interleukin-10,IL-10)、白细胞介素-17(interleukin 17,IL-17)和干扰素-γ(INF-γ)的影响。方法将48只SPF级雄性大鼠随机分为正常组、模型组、柴芍六君汤组和美沙拉嗪组。采用葡聚糖硫酸钠(DSS)+束缚法+饮食失节法建立肝郁脾虚型UC大鼠模型,采用高架十字迷宫实验(Elevated plus-maze test,EPM)、旷场实验(Open field test,OFT)和新环境进食抑制实验(Novelty suppressed feeding test,NSF)对模型进行评价。正常组和模型组给予等量生理盐水灌胃,柴芍六君汤组给予柴芍六君汤灌胃,用量为20.25 g/kg;美沙拉嗪组给予美沙拉嗪灌胃,用量为0.35 kg。采用ELISA和RT-PCR检测结肠黏膜IL-4、IL-10、IL-17、INF-γ指标的表达量。结果采用葡聚糖硫酸钠+束缚法+饮食失节法能够成功建立肝郁脾虚型UC动物模型。ELISA和RT-PCR检测结果显示,与正常组比较,模型组大鼠IL-4和IL-10含量明显降低,而IL-17和IFN-γ含量明显升高,差异有统计学意义(P<0.05);与模型组比较,柴芍六君汤组和美沙拉嗪组IL-17和IFN-γ降低趋势明显(P<0.05),而IL-4和IL-10呈现升高趋势(P<0.05)。结论葡聚糖硫酸钠+束缚法+饮食失节法能够成功建立肝郁脾虚型UC大鼠模型,柴芍六君汤能够通过调节细胞炎性因子的表达量而参与对UC的治疗作用,从而发挥对T细胞介导的炎性平衡的调节作用,减少炎性反应,改善肠道功能。Objective To establish and evaluate an animal model of ulcerative colitis(UC)with liver-depression and spleen-deficiency syndrome,and to analyze the effect of Chaishao Liujun Decoction(CSLJD)on the gene and protein expression of inflammatory response factors including L-4、IL-10、IL-17 and INF-γ.Methods Forty-eight SPF rats were randomly divided into 4 groups:normal group,model group,CSLJD treatment group and mesalazine treatment group.The normal group received no intervenes;the other groups underwent take orally with dextran sulphate sodium salt(DSS)+tight-binding+improper diet to establish a model of UCwith liver-depression and spleen-deficiency syndrome.After the establishment,the model was evaluated by using open field test,elevated plus-maze test and feeding inhibition test in new environment.The normal group and the model group were given the same amount of normal saline by gavage.The CSLJD treatment group and mesalazine treatment group were treated with 20.25 g/kg CSLJTD and 0.35 g/kg mesalazine,respectively.One week after treatment,the expression of IL-4,IL-10,IL-17 and INF-γin the colonic mucosa were detected by ELISA and RT-PCR.Results Using dextran sodium sulfate+restraint method+eating disorder method can successfully establish a UCanimal model of liver depression and spleen deficiency.The results of ELISA and RT-PCR showed that compared with the normal group,the levels of IL-4 and IL-10 in the model group were significantly reduced,while the levels of IL-17 and IFN-γwere significantly increased,and the difference was statistically significant(P<0.05);Compared with the model group,the IL-17 and IFN-γof the Chaishao Liujun decoction group and the mesalazine group decreased significantly(P<0.05),while IL-4 and IL-10 showed an increasing trend(P<0.05).Conclusion Dextran sodium sulfate+restraint method+diet disorder method can successfully establish a UCrat model of liver depression and spleen deficiency.In addition,CSLJD can participate in the treatment of UC through regulating the expression of in
