摘  要:目的分析瑞舒伐他汀对脑梗死大鼠神经细胞保护的作用机制。方法通过大脑中动脉闭塞成功建立30只脑梗死模型大鼠,将其采用随机数字表法分为3组,每组各10只,分别为模型组(每日灌注等量0.9%氯化钠溶液),低剂量组(每日腹腔注射2.5μmol/L的瑞舒伐他汀),高剂量组(每日腹腔注射40μmol/L的瑞舒伐他汀),并随机纳入10只健康大鼠为健康对照组(每日灌注等量0.9%氯化钠溶液)。比较各组血清白细胞介素(IL)-6、IL-8、IL-1β、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)水平,并比较各组UCP2、SIRT3及其mRNA水平和脑神经元细胞增殖、凋亡、侵袭和迁移情况。结果与健康对照组比较,模型组大鼠血清IL-6、IL-8、IL-1β、TNF-α和MDA、CAT、GSH-PX水平均显著升高,血清SOD水平显著降低,差异均有统计学意义(P<0.05);与模型组比较,低剂量组和高剂量组大鼠血清IL-6、IL-8、IL-1β、TNF-α和MDA、CAT、GSH-PX水平均显著降低,血清SOD水平显著升高,差异均有统计学意义(P<0.05)。与健康对照组比较,模型组大鼠脑组织UCP2、SIRT3及其mRNA水平均显著升高,差异均有统计学意义(P<0.05);与模型组比较,低剂量组和高剂量组大鼠脑组织UCP2、SIRT3及其mRNA水平均显著降低,差异均有统计学意义(P<0.05)。与健康对照组比较,模型组大鼠脑神经元细胞A值显著降低,细胞凋亡率、培养24、48、72 h时脑神经元细胞侵袭个数、细胞迁移率均显著升高,差异均有统计学意义(P<0.05);与模型组比较,低剂量组和高剂量组大鼠脑神经元细胞A值显著升高,细胞凋亡率、培养24、48、72 h时脑神经元细胞侵袭个数、细胞迁移率均显著降低,且高剂量组以上指标较低剂量组变化更显著,差异均有统计学意义(P<0.05)。结论瑞舒伐他汀可通过调控UCP2-SIRT3信号通路而抑制脑梗死大鼠神经细胞凋亡,Objective To analyze the protective mechanism of rosuvastatin on nerve cells in rats with cerebral infarction.Methods Thirty cerebral infarction model rats were successfully established through middle cerebral artery occlusion,and they were divided into 3 groups by random number table method,each with 10 rats,respectively,the model group(perfusion of the same amount of 0.9%sodium chloride solution daily)and the low-dose group(each daily intraperitoneal injection of 2.5μmol/L rosuvastatin),high-dose group(intraperitoneal injection of 40μmol/L rosuvastatin daily),and randomly included 10 healthy rats as healthy control group(perfusion of the same amount of 0.9%sodium chloride solution daily).The levels of serum interleukin(IL)-6,IL-8,IL-1β,tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT)were compared,and glutathione peroxidase(GSH-PX),and the levels of UCP2,SIRT3 and their mRNA,the proliferation,apoptosis,invasion and migration of brain neurons were compared.Results Compared with the healthy control group,the serum levels of IL-6,IL-8,IL-1β,TNF-αand MDA,CAT,GSH-PX in the model group were significantly increased,and the serum SOD levels were significantly decreased,and the differences were statistically significant significance(P<0.05);compared with the model group,the levels of serum IL-6,IL-8,IL-1β,TNF-αand MDA,CAT,GSH-PX in the low-dose and high-dose groups were significantly decreased,the level of serum SOD was significantly increased,and the differences were statistically significant(P<0.05).Compared with the healthy control group,the levels of brain tissue UCP2,SIRT3 and mRNA in the model group were significantly increased,and the differences were statistically significant(P<0.05);compared with the model group,the levels of brain tissue UCP2,SIRT3 and mRNA in the low-dose and high-dose groups were significantly decreased,and the differences were statistically significant(P<0.05).Compared with the healthy control group,the A value of rat brain neuron cells in
