摘  要:试验旨在探究不同添加剂量的稀土壳糖胺螯合盐(rare earth-chitosan chelate,RECC)对肉仔鸡脾脏免疫和抗氧化功能的影响。选取1日龄爱拔益加(AA)肉仔鸡192只,随机分为4组,每组6个重复,每个重复8只。对照组肉仔鸡饲喂基础日粮,试验组分别在基础日粮中添加150、200、250 mg/kg的RECC。试验期42 d。结果显示,随着RECC添加剂量的增加,肉仔鸡脾脏中白细胞介素-2 (IL-2)、白细胞介素-4 (IL-4)、免疫球蛋白G (IgG)、免疫球蛋白A (IgA)、免疫球蛋白M (IgM)和谷胱甘肽过氧化物酶(GSH-Px)活性、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性增加,白细胞介素-1β(IL-1β)、白细胞介素-6 (IL-6)的含量降低。试验前期、后期,TLR4/NF-κB途径中细胞因子的基因表达量降低,TLR3表达量有所上升;试验后期,Nrf2和GPx-7表达量升高效应。研究表明,日粮中添加150~200 mg/kg RECC可提高肉仔鸡脾脏的抗炎因子和免疫球蛋白含量,降低促炎因子含量,增强抗氧化酶活性,抑制TLR4/NF-κB信号通路,激活TLR3和Nrf2通路,提高抗氧化因子GPx-7的表达水平,且与RECC添加量存在剂量依赖关系。The experiment was to investigate the effect of different doses of rare earth-chitosan chitosan(RECC) on spleen immunity and antioxidant function of broilers. A total of 192 one-day-old Arbor Acres(AA) broilers were randomly divided into four groups, with six replicates in each group and each broilers in each replicate. Broilers in control group were fed with basal diet, and broilers in experimental groups were fed with experimental diets supplemented with150, 200 and 250 mg/kg RECC, respectively. The experiment lasted for 42 d. The results showed that as the added dose of RECC increased, IL-2, IL-4, IgG, IgA, IgM, GSH-Px, T-AOC and SOD in spleen of broilers were increased. The content of IL-1β and IL-6 decreased. Before and after the experiment, the gene expression of TLR4/NF-κB pathway decreased, while the expression of TLR3 increased. The expression levels of Nrf2 and GPX-7 increased in later stage. The experiment indicates that adding 150~200 mg/kg RECC to the diet can increase the content of anti-inflammatory factors and immunoglobulins in spleen of broilers, reduce the content of pro-inflammatory factors, enhance the activity of antioxidant enzymes, inhibit the TLR4/NF-κB signaling pathway, activate the TLR3 and Nrf2 pathways, increase the expression level of antioxidant factor GPx-7. There is a dose-dependent relationship with the addingamount of RECC.
