催奶灵散对子鼠生长性能和肠道菌群结构的影响

摘  要:试验采用16S rRNA测序分析摄入催奶灵散泌乳母鼠及其子鼠盲肠微生物菌群结构和多样性差异,检测子鼠生长性能、母鼠及其子代免疫性能和抗氧化性能,探讨催奶灵散对子代生长发育的影响。选取产仔时间相差不超过1 d、体重(55.00±2.00) g的产后母鼠20只,并调整至每只母鼠带仔鼠10只,随机分为试验组和对照组。试验组母鼠剂量灌胃催奶灵散1 mL/(只·d),对照组母鼠灌胃同等剂量的生理盐水。结果显示,与对照组相比,试验组摄入催奶灵散母鼠乳腺总抗氧化能力极显著提高(P<0.01),子鼠1 w的平均体重极显著提高(P<0.01),第2 w、3 w的平均体重和平均日增重极显著下降(P<0.01)。试验组母鼠Biomarker菌群为拟杆菌门拟杆菌纲拟杆菌目副拟杆菌属、Tannerellaceae和Actinobacteria,肠道菌群倾向于尿素分解代谢和硝酸盐代谢;试验子鼠Biomarker菌群梭状芽孢杆菌目Clostridia、放线菌门、变形菌门,肠道中与硝酸盐代谢相关微生物以及与病原性腹泻等相关的菌群显著高于对照组(P<0.05)。研究表明,催奶灵散能够影响泌乳母鼠肠道菌群结构的多样性和子代生长发育和肠道菌群结构。The study analyzed the cecal microflora structure and diversity of suckling lactating rats and the progeny by16 S rRNA sequencing. Growth performance, immune performance and antioxidant performance of the progeny were detected, and the effect of Cuinailing on growth and development of progeny were investigated. Twenty postpartum female rats with a body weight of(55.00±2.00) g and 1 day delivery interval were selected and adjusted to 10 offspring per female rat. The rats were randomly divided into experimental group and control group. Each female rat in experimental group was given 1 m L/d, and the control group was given the same dose of normal saline. The results showed that compared with the control group, the total antioxidant capacity of mammary gland of Cuinailing in experimental group was extremely increased(P<0.01), the average body weight at 1 w was extremely increased(P<0.01), and the average body weight and average daily gain at 2 w and 3 w were extremely decreased(P<0.01). The Biomarker flora of the experimental group were Parabacteroidetes, Tannerellaceae and Actinobacteria, and the intestinal flora tended to urea catabolism and nitrate metabolism. Clostridia, Actinomycetes, Proteobacteria, and bacteria related to nitrate metabolism and pathogenic diarrhea were significantly higher than those in the control group(P<0.05). The experiment indicates that Cuinailing can affect the diversity of intestinal microflora structure and the growth and development of progeny and intestinal microflora structure in lactating mice.
