摘  要:目的基于UPLC定量测定与响应曲面法从多指标和综合评价角度优选"建昌帮"姜天麻提取工艺。方法采用UPLC定量分析,色谱柱为Acquity UPLC BEH C;柱(100 mm×2.1 mm,1.7 μm),体积流量0.2 mL·min^(-1),柱温35℃,以流动相为乙腈(A)-0.05%磷酸水溶液(B),梯度洗脱(0~14 min,3%~29%A);检测波长为220 nm。以天麻素,天麻苷元,对羟基苯甲酸,巴利森苷A、B、C、E及综合评分为评价指标,在考察料液比、提取时间、提取温度、粒径、提取溶剂等单因素的基础上,运用响应曲面设计法对药效成分影响较大的料液比、提取温度及提取溶剂之间的交互作用进行考察,并优选出姜天麻最佳提取工艺。结果 姜天麻最佳提取工艺:70%乙醇,料液比1:15,80℃超声40 min。结论优选出的姜天麻提取工艺稳定可行,可为"建昌帮"姜天麻深入研究及临床应用提供理论依据。Objective To determine and optimize the extraction technology of Gastrodia elata processed with ginger juice by response surface methodology and UPLC method.Methods The UPLC quantitative analysis was used.The column was Acquity UPLC BEH C;column (100 mm×2.1 mm,1.7 μm) with a volume flow rate of 0.2 mL·min^(-1) and a column temperature of 35℃.The evaluation index:the mobile phase was acetonitrile (A)-0.05% phosphoric acid aqueous solution (B),with gradient elution (0-14 min,3%-29%A).The detection wavelength was 220 nm.Gastrodin,gastrodigenin,p-hydroxybenzoic acid,parishin A,B,C,E and comprehensive score were used as the evaluation indexes.Response surface design was used to determine the effect of interaction among solid-liquid ratio,extraction temperature and extraction solvent on the extraction process of Gastrodia elata based on the single factor of solid-liquid ratio,extraction time,extraction temperature,particle size and extraction solvent.Results The best extracted technology of Gastrodia elata was 70% ethanol as extraction solvent,the ratio of material to liquid is 1∶15,and the extraction temperature was 80℃ for 40 min by ultrasonic extraction.Conclusion The optimized extraction process of Gastrodia elata is stable and feasible,which provides scientific basis for the formulation of quality standards and research and application of Gastrodia elata.
