摘  要:目的:建立养血生发合剂HPLC指纹图谱并结合化学模式识别,对养血生发合剂整体质量进行评价。方法:采用Waters XBridge C_(18)色谱柱(250 mm×4.6 mm, 5μm),以甲醇-0.2%磷酸水为流动相,梯度洗脱,流速:1.0 ml·min^(-1),柱温:30℃,并通过层次聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘回归分析(OPLS-DA)对养血生发合剂整体质量进行分析。结果:建立了养血生发合剂指纹图谱,共标定17个共有峰,指认了其中7个共有峰。方法学考察结果表明,所建立的指纹图谱方法精密度、稳定性和重复性均良好,简便可靠。10批养血生发合剂相似度均≥0.977,可聚为3类,2,3,5,4’-四羟基二苯乙烯-2-O-β-D葡萄糖苷、芍药苷和没食子酸是导致不同批次样品间差异的标志性成分。结论:指纹图谱和化学模式识别相结合的考察模式准确可行,可用于养血生发合剂的质量评价。Objective: To establish the HPLC fingerprints of Yangxue Shengfa mixture and evaluate its quality combined with chemical pattern recognition. Methods: The method was developed on a column of Waters XBridge C_(18)(250 mm×4.6 mm, 5 μm) with methanol-0.2% phosphoric acid solution as the mobile phase with gradient elution at the flow rate of 1.0 ml·min^(-1). The column temperature was 30℃. The quality of Yangxue Shengfa mixture was evaluated by HCA, PCA and OPLS-DA. Results: The characteristic fingerprints of Yangxue Shengfa mixture was established. A total of seventeen common peaks were calibrated and seven common peaks were identified. The results of methodological investigation showed that the established fingerprint method had good precision, stability and repeatability. The method was simple and reliable. The similarity of Yangxue Shengfa mixture was greater than or equal to 0.977, which could be grouped into three categories, and 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside, paeoniflorin and gallic acid were the markers leading to the differences among different batches of samples. Conclusion: The combination of fingerprint and chemical pattern recognition is accurate and feasible. It can be applied to the quality evaluation of Yangxue Shengfa mixture.
