摘 要:目的优选斑龙强志膏滋3味药材(刺五加、林蛙油、鹿角胶)的最佳配伍比例,并研究其对免疫功能低下小鼠的免疫恢复作用。方法采用星点-效应面法(CCD-RSM),以小鼠血清中免疫球蛋白含量(IgA、IgG、IgM)为指标,确定复方斑龙强志膏滋各药材的最佳配比。最佳配比按照前期提取工艺制备药液,并以此为高剂量组,另0.5倍、0.25倍稀释成中、低剂量组。150只小鼠随机分为5批,每批30只,分为空白组、模型组、斑龙强志膏滋高、中、低剂量组。环磷酰胺建立免疫低下模型,通过体质量、脏器指数、体外细胞实验如:检测T淋巴细胞的增殖能力、巨噬细胞吞噬能力、抗体生成细胞数、NK细胞活性确定最佳用药剂量。结果斑龙强志膏滋高、中、低剂量组均可显著提高小鼠的体质量、脏器指数、T淋巴细胞的增殖、巨噬细胞吞噬能力、抗体生成细胞数、NK细胞活性,尤其高剂量组最为显著。结论斑龙强志膏滋可有效改善环磷酰胺所致免疫低下小鼠的免疫状态。Objective To screen the best compatibility ratio of Banlongqiangzhi Gaozi 3 herbs(Acanthopanax, Rhizoma Oleum and Antler Gum), and to study the immune recovery effect of Banlongqiangzhi on immunocompromised mice.Methods The optimal ratio of compound Banlongqiangzhi ointment was determined by using the central point-response surface method with the contents of immunoglobulin(IgA, IgG, IgM) in serum of mice as indexes.The optimal proportion of the liquid was prepared according to the extraction process in the early stage, and the liquid was used as the high-dose group, and the other 0.5 times and 0.25 times were diluted into the medium-dose and low-dose groups.150 mice were randomly divided into 5 batches with 30 mice in each batch, and they were divided into blank group, model group, Banlongqiangzhi ointment high-dose, medium-dose and low-dose groups.The immunodeficiency model was established by cyclophosphamide. The content of immunoglobulin(IgA, IgG, IgM) in serum was determined by body weight, organ index, ELISA, and in vitro cell experiments such as T lymphocyte proliferation, macrophage phagocytosis, number of antibody-producing cells, and NK cell activity were determined to determine the optimal dosage.Results The body weight, organ index, immunoglobulin level, T lymphocyte proliferation, macrophage phagocytosis, number of antibody-producing cells and NK cell activity of mice in high, medium and low dose groups were significantly increased, especially in high dose group.Conclusion Banlongqiangzhi ointment can effectively improve the immune state of immunocompromised mice induced by cyclophosphamide.
