摘 要:非酒精性脂肪性肝病(Non-alcoholic fatty liver disease,NAFLD)是指肝细胞内以脂肪变性为主要特征的临床病理综合征,而资料显示阿育魏实具有一定的降脂活性,可以改善NAFLD。因此本研究采用Box-Benhnken响应面法筛选阿育魏实挥发性成分(Trachyspermum ammi(L.)Sprague volatile constituent,TSVC)提取参数并初步考察其体外活性研究。在单因素基础上,采用四因素三水平响应面法筛选阿育魏实挥发性成分的提取工艺参数,分别建立NAFLD和胰岛素抵抗细胞模型并进行干预,微量法检测细胞葡萄糖消耗量、TG、TC、AST、ALT,进行油红染色考察其改善非酒精性脂肪肝和胰岛素抵抗活性。当TSVC最佳提取条件为料液比9mL/g、浸泡时间12h、超声时间15min和提取时间6.5h,其提取率为2.12%;体外实验结果表明:与模型组相比,阿育魏实挥发性成分中、高剂量组降低细胞内的TG、TC、AST、ALT含量;增加胰岛素抵抗模型细胞的葡萄糖消耗量;油红染色结果中看出可以减少脂质沉积。总之,阿育魏实挥发性成分在一定浓度范围内可改善非酒精性脂肪肝和胰岛素抵抗,为其后期研究提供理论依据和实验基础。non-alcoholic fatty liver disease(NAFLD)is a clinicopathological syndrome characterized by fatty degeneration in hepatocytes. The data showed that ayuweishi has certain lipid-lowering activity and can improve NAFLD. Therefore,in this study,the box-Benhnken response surface method was used to select the extraction parameters of Trachyspermum ammi(L.)Sprague volatile constituent(TSVC)and to study its in vitro activity.Based on single factor,the four factors three levels of response surface method was used to filter the ayuweishi technological parameters of extracting volatile components,respectively established NAFLD and insulin resistance model and intervene to trace method to detect cell glucose consumption,TG,TC,AST,and ALT,and oil red staining method was used to study the improvement of nonalcoholic fatty liver disease and the activity of insulin resistance. When the optimal extraction conditions of TSVC were solid-liquid ratio of 9 mL/g,immersion time of 12 h,ultrasonic time of 15 min and extraction time of 6.5 h,the extraction rate was 2.12%. The results of in vitro experiments showed that compared with the model group,the contents of TG,TC,AST and ALT in cells of the medium and high dose groups of volatile components of Trachyspermum ammi(L.)Sprague decreased,and the glucose consumption of the insulin resistant model cells increased. The results of oil red staining showed that the lipid deposition could be reduced. In conclusion,the volatile components of Ayuwei can improve nonalcoholic fatty liver disease and insulin resistance in a certain concentration range,providing theoretical basis and experimental basis for the later research.
