摘 要:目的探究白细胞分化抗原83(cluster of differentiation 83,CD83)和热休克转录因子5(heat shocktranscription factor 5,HSF5)鉴别生前、死后皮肤烧伤的法医学应用价值。方法通过逆转录实时定量聚合酶链反应(reverse transcription real-time quantitative polymerase chain reaction,RT-q PCR),定量检测生前及死后烧伤小鼠皮肤组织和人类皮肤组织中CD83和HSF5 m RNA的表达情况。结果与空白对照和死后烧伤组相比,生前烧伤组小鼠CD83和HSF5的m RNA水平明显升高,且死后96 h可检测CD83 m RNA高表达,死后72 h可检测HSF5 m RNA表达。15例人类烧伤皮肤和未烧伤自身皮肤相比,11例样本CD83和HSF5的m RNA水平在烧伤处升高(P<0.05),4例没有变化。结论CD83和HSP5有望作为生前烧伤鉴定的生活反应指标应用于法医学实践。Objective To explore the forensic application value of cluster of differentiation 83(CD83)and heat shock transcription factor 5(HSF5)in identifying antemortem and postmortem skin burns.Methods Through reverse transcription real-time quantitative polymerase chain reaction(RT-qPCR),CD83 and HSF5 mRNA levels in the skin tissues of antemortem and postmortem burned mice and hu-man samples were detected quantitatively.Results Compared with the control group and the postmor-tem burned group,the mRNA levels of CD83 and HSF5 in antemortem burned mice were higher.The high mRNA expressions of CD83 could be detected 96 h after death,and the mRNA expressions of HSF5 could be observed 72 h after death.Compared with undamaged skin,increased CD83 and HSF5 mRNA levels were detected in 11 out of 15 cases(P<0.05).Conclusion CD83 and HSF5 can be used in forensic practice as indicators for vital reaction in antemortem burn identification.
