摘 要:GRF是植物特有的转录因子,主要参与调控植物生长发育过程。本研究利用生物信息方法对蓖麻GRF (RcGRF)基因进行全基因组鉴定,并对其理化性质、基因结构、保守结构域、系统发育、组织表达分析以及外源赤霉素(GA)和多效唑(PAC)处理两个蓖麻品种(高杆‘滇蓖2号’和矮杆‘滇蓖5号’)后顶端嫩茎转录组测序分析。结果表明,蓖麻共有9个GRF转录因子,蛋白长度233~617 aa,等电点(pI)为6.34~9.48,每个成员含有2~4个内含子,每个RcGRF的蛋白结构域均包括一个由39~43个氨基酸组成的WRC保守结构域和一个由34个氨基酸组成QLQ保守结构域。系统发育分析表明蓖麻与拟南芥GRF的亲缘关系比水稻更近。不同组织表达结果表明多数RcGRF基因在发育的胚乳及萌发的种子中高表达,而RcGRF2在叶片、雄花和萌发的种子中都不表达。GA和PAC处理转录组测序结果显示GA处理DB2 RcGRF1、DB2 RcGRF17下调表达,GA处理DB5 RcGRF1、DB5 RcGRF4下调表达,PAC处理DB2 RcGRF4、DB2 RcGRF6上调表达、RcGRF9下调表达,表明RcGRF1、RcGRF4、RcGRF7、RcGRF9可能通过赤霉素途径调节植物株型。本研究为蓖麻GRF基因对生长发育调控机理研究奠定分子基础,为调控蓖麻株高相关基因的研究提供科学依据。GRF is a plant specific transcription factor which is mainly involved in the regulation of plant growth and development.In this study,the castor GRFs(RcGRFs) in the whole genome were identified by bioinformatics,and their physicochemical properties,gene structure,conserved domain,phylogeny and tissue expression were analyzed.In addition,exogenous gibberellin(GA) and paclobutrazol(PAC) were used to treat castor cultivar tall DB2 and dwarf DB5,and then transcriptome sequencing was performed on apical tender stem.There were 9 GRF transcription factors in castor,with the protein length from 233 to 617 aa,the isoelectric point from 6.34 to 9.48,and the intron number from 2 to 4.The protein domain of each RcGRF includes a WRC conserved domain composed of 39~43 aa and a QLQ conserved domain composed of 34 aa.Phylogenetic analysis showed that the relationship between castor and Arabidopsis GRF was closer than that of rice.The results of different tissues showed that most RcGRF genes were highly expressed in developing endosperm and germinating seeds,while RcGRF2 was not expressed in leaves,male flowers and germinating seeds.GA and PAC treatment transcriptome sequencing results showed RcGRF1 and 7 are down-regulated after GA processing DB2,RcGRF1 and DB2 RcGRF4 were down-regulated after DB5 treatment by GA,the up-regulated expression of DB2 RcGRF4 and DB2 RcGRF6 and down-regulated expression of RcGRF9 after PAC processing DB2.It suggests that RcGRF1,RcGRF4,RcGRF7 and RcGRF9 May regulate plant type through the gibberellin pathway.This study will lay a molecular foundation for the regulation mechanism study of castor GRF gene on growth and development,and it provides scientific basis for the regulation of height-related genes in castor.
