摘 要:沉香(Agarwood)是一种名贵中药和天然香料,倍半萜是其主要的特征性成分之一。法呢基焦磷酸合酶FPS(Farnesyl pyrophosphate synthase)是萜类化合物生物合成的关键酶之一。本研究根据转录组数据在白木香中克隆一个新的法呢基焦磷酸合酶基因AsFPS2,该基因全长1432 bp,包含一个1287 bp的完整开放阅读框,编码428个氨基酸。As FPS2具有典型的植物法呢基焦磷酸合酶结构特征包含5个保守的结构域和2个保守的DDXXD基序,与小果沉香(Aquilaria microcarpa)中的法呢基二磷酸合酶蛋白具有86.55%的同源性。AsFPS2基因编码区包含12个外显子和11个内含子。AsFPS2启动子区含有多个应答激素和胁迫的响应元件。表达分析显示,伤害、茉莉酸甲酯和乙烯利处理能显著诱导AsFPS2的表达。研究结果说明AsFPS2可能参与了沉香倍半萜的生物合成,为进一步揭示其在倍半萜积累和沉香形成中的作用打下基础。Agarwood is aprecious traditional Chinese medicine and natural flavor.Sesquiterpenes are the main chemical characteristic constituents of agarwood.Farnesyl pyrophosphate synthase(FPS) is one of the key enzymes in terpenoid biosynthesis.In this study,a novel farnesyl pyrophosphate synthase gene named AsFPS2 was cloned in Aquilaria sinensis based on transcriptome data.The full length of AsFPS2 is 1 432 bp with a 1 353 bp opening reading frame(ORF) encoding 428 amino acids.As FPS2 has the typical structural characteristics of plant farnylpyrophosphate synthase and contains five conserved domains and two conserved DDXXD motifs,which share high homology with similar FPS proteins in other plants.The AsFPS2 gene contains 12 exons and 11 introns.Several responsive elements in response to hormones and stress are found in the promoter region of AsFPS2.Expression analysis showed that AsFPS2 was significantly induce by wounding,methyl jasmonate and ethephon treatment.The results showed that AsFPS2 may be involved in the sesquiterpene biosynthesis,which lays a foundation for further revealing its role in the accumulation of sesquiterpenes and agarwood formation.
