摘 要:本研究采用响应曲面法、MTT比色法分别研究蒲公英根多糖的硫酸酯化工艺与蒲公英根多糖及其硫酸酯对H_(2)O_(2)诱导的HepaRG细胞氧化损伤的保护作用。结果表明:蒲公英根多糖硫酸酯化反应的最优条件为反应温度83℃、吡啶与氯磺酸比为8、反应时间3 h。蒲公英根多糖及其硫酸酯对H_(2)O_(2)诱导的HepaRG细胞氧化损伤均具有保护作用,而且硫酸酯化蒲公英根多糖的活性优于蒲公英根多糖。In this study,response surface methodology(RSM)and MTT colorimetry were used to study the sulfation process of dandelion root polysaccharide and the protective effects of dandelion root polysaccharide and its sulfation on oxidative damage of HepaRG cells induced by hydrogen peroxide(H_(2)O_(2)).The results showed that the optimum conditions for sulfation of dandelion root polysaccharides were as follows:reaction temperature was 83℃,ratio of pyridine to chlorosulfonic acid was 8,reaction time was 3 h.Moreover,dandelion root polysaccharide and its sulfates had protective effects on H_(2)O_(2)induced oxidative damage of HepaRG cells,and the activity of sulfated dandelion root polysaccharide was better than that of dandelion root polysaccharide.
