摘 要:目的评估不同培养基对产生物膜白假丝酵母菌芽管产生的影响。方法收集临床产生物膜的白假丝酵母菌20株,购买3株可以产生物膜的质控菌株。比较23株白假丝酵母菌在5种不同培养基(人血清、胰蛋白胨大豆肉汤、脑心浸液肉汤、RPMI 1640、NaHCO_(3)溶液)的出芽菌株数量和出芽率。结果在23株产生物膜的白假丝酵母菌中,人血清和脑心浸液肉汤培养基中23株(100.0%)全部产生芽管,NaHCO_(3)培养基18株(78.3%)、胰蛋白胨大豆肉汤培养基16株(69.6%)和RPMI 1640培养基15株(65.2%)。芽管生成试验阳性的白假丝酵母菌中,在5%CO_(2)培养条件下的酵母细胞出芽率更高(孵育2h,出芽率78.0%),芽管长度更长。结论脑心浸液肉汤可以代替人血清作为芽管试验的诱导剂,人血清(5%CO_(2))可以促进芽管的生成以及延伸芽管的长度。Objective To evaluate the effect of different liquid media inducing germ tube formation in Candida albicans.Methods A total of 20strains of biofilm-producing Candida albicans were clinically collected.Standard control strains were purchased.The numbers and germ tube formation rates of these Candida albicans strains in 5different liquid media(human serum,tryptone soy broth,brain heart infusion broth,RPMI 1640,NaHCO_(3) solution)were compared.Results Among the 5liquid media,human serum and brain heart infusion broth gave 100.0%germ tube production,NaHCO_(3) solution gave 78.3%,tryptone soy broth gave 69.6%,and RPMI 1640gave 65.2%germ tube production at the end of 3hours of incubation.In the Candida albicans strains with positive results of germ tube formation experiment,the germination rate of yeast cells was higher in 5%CO_(2)(2hours,78.0%),and the germ tube was longer.Conclusion Brain heart infusion broth can replace human serum for inducing germ tube production of Candida albicans;and human serum(5%CO_(2))can promote the formation and increase the length of germ tube.
