摘　　要：目的研究鼠肉瘤病毒家族相关蛋白2A(RAB2A)在胃癌组织样本中的表达以及对胃癌细胞增殖、侵袭和迁移能力的影响。方法 (1)收集胃癌患者,将胃癌组织分为癌组织组及癌旁组织组,用荧光定量-PCR法检测RAB2A基因表达水平。(2)将MGC-803细胞分为4组:过表达对照组、过表达RAB2A组、干扰表达对照组和干扰表达RAB2A组(分别命名为第1、第2、第3和第4转染组)。用慢病毒感染细胞,第1转染组感染对照慢病毒,第2转染组感染过表达RAB2A慢病毒,第3转染组感染干扰对照慢病毒,第4转染组感染干扰表达RAB2A慢病毒,各组的感染量根据细胞数目决定,感染复数均为5(细胞与病毒颗粒比为1∶5),处理2 d。用噻唑蓝法检测细胞增殖水平(光密度)值,用划痕实验检测细胞迁移情况,用侵袭实验检测细胞侵袭数量,用蛋白质印迹法检测RAB2A和细胞外调节蛋白激酶(Erk)信号通路相关蛋白的蛋白表达水平。结果 (1)RAB2A在正常组织和癌组织中的相对表达水平分别为0.66±0.39和1.04±0.59,癌组织中RAB2A表达水平为正常组织的1.58倍。(2)第1、第2、第3和第4转染组的伤口愈合率为(50.91±4.33)%,(81.03±6.21)%,(73.97±2.55)%和(37.99±4.83)%;这4组的穿孔细胞数分别为67.67±8.32,135.67±16.17,75.00±10.54和43.00±12.29;这4组的磷酸化Erk相对表达量分别为0.97±0.02,1.51±0.12,1.14±0.11,0.46±0.13。第2转染组的上述指标与第1转染组比较,或第4转染组的上述指标和第3转染组比较,差异均有统计学意义(均P<0.01)。结论抑制RAB2A表达可以抑制胃癌细胞的增殖、侵袭和迁移能力,而过表达RAB2A则能促进胃癌细胞的增殖、侵袭和迁移。Objective To study the expression of murine sarcoma virus family related protein 2 A(RAB2 A) in gastric cancer tissue samples and its effect on the proliferation, invasion and migration of gastric cancer cells. Methods(1)Gastric cancer patients were collected and divided into two groups: cancer tissues group and adjacent tissues group, and fluorescence quantitative-PCR was used to detect the expression level of RAB2 A. The expression level of RAB2 A in cancer tissues is 1.58 times that of normal tissues.(2)The MGC-803 cells were divided into 4 groups: overexpression control group, overexpression RAB2 A group, interference expression control group, and interference expression RAB2 A group(named transfection-1 group, transfection-2 group transfection-3 group,and transfection-4 group). In each group treatment,the amount of infection is determined by the number of cells,and the multiplicity of infection is all 5( the ratio of cells to virus particles is 1 ∶ 5) and the treatment duration was 2 days. The RNA expression level was verified using quantitative-PCR;the proliferation rate of cells was detected using diphenyl tetrazolium bromide( MTT) method;cell migration was detected using wound healing assay;cell invasion was detected using invasion test;the effect of RAB2 A expression on extracellular signal-regulated kinase( Erk) signaling pathway was detected using western blot. Results( 1) The relative expression level of RAB2 A in normal tissues and cancer tissues were 0. 66 ± 0. 39 and 1. 04 ± 0. 59,respectively;the expression level of RAB2 A in cancer tissues was 1. 58 times higher than that in normal tissues.( 2) The wound healing rates in transfection-1 group,transfection-2 group,transfection-3 group,and transfection-4 group were( 50. 91 ± 4. 33) %,( 81. 03 ± 6. 21) %,( 73. 97 ± 2. 55) % and( 37. 99 ± 4. 83) %;the number of perforated cells in the 4 groups were 67. 67 ± 8. 32,135. 67 ± 16. 17,75. 00 ± 10. 54 and 43. 00 ± 12. 29,respectively;the relative expression levels of phosphorylated Erk in the 4
