凉血消风汤对血热型银屑病患者外周血单核细胞NF-κB信号通路表达的影响

摘　　要：目的探讨凉血消风汤对血热型银屑病患者外周血单个核细胞(PBMC)中核因子(NF)-κB通路表达的影响。方法设立正常对照组和银屑病组,银屑病组给予凉血消风汤治疗4周。记录治疗前后银屑病患者银屑病皮损面积与严重性指数(PASI)评分以观察临床疗效。采集正常组及银屑病组治疗前后外周血,经密度梯度离心法获得PBMC。用实时荧光定量聚合酶链反应(RT-PCR)方法测定PBMC中NF-κB通路蛋白IKKβ、IκBα、p50、p65的基因表达水平。用蛋白质印迹法(Western blot)检测PBMC中IKKβ、p-IκBα、p50、p-p65的蛋白表达水平。结果银屑病患者经凉血消风汤治疗前后比较,PASI评分降低(P<0.05);与正常对照组比较,银屑病组治疗前PBMC中NF-κB相关蛋白的基因和蛋白表达水平均升高(P<0.05),与银屑病组治疗前比较,银屑病组治疗后NF-κB相关蛋白的基因和蛋白表达水平均明显降低(P<0.05),差异有统计学意义。结论凉血消风汤能降低血热型银屑病患者PBMC中NF-κB通路蛋白的基因和蛋白表达水平,抑制NF-κB通路的活化,是其发挥治疗银屑病作用的机制之一。Objective In order to investigate the effects of Liangxue Xiaofeng Tang on expression of nuclear factor kappa-B(NF-κB)signaling pathway in peripheral blood mononuclear cell(PBMC)of blood-heat psoriasis.Methods The normal control group and the psoriasis group were established.The psoriasis group was given Liangxue Xiaofeng Tang for four weeks.The psoriasis area and severity index(PASI)score of patients with psoriasis before and after treatment were recorded to observe the clinical efficacy.Peripheral blood was collected before and after treatment,normal blood was collected from peripheral blood,and PBMC was obtained by density gradient centrifugation.The gene expression level of NF-κB related proteins inhibitor kappa B kinaseβ(IKKβ),inhibitor kappa Bα(IκBα),p50,and p65 in PBMC was measured by real-time fluorescence quantitative polymerase chain reaction(PCR).Western blotting was used to detect the protein expression level of IKKβ,p-IκBα,p50and p-p65 in PBMCs.Results The PASI scores was decreased significantly by taking Liangxue Xiaofeng Tang after the treatment(P<0.05).Compared with the normal control group,the gene and protein expression levels of NF-κB related protein in PBMC before treatment in the psoriasis group were increased(P<0.05).Compared with the psoriasis group,the gene and protein expression levels of NF-κB related proteins were significantly decreased after treatment in the psoriasis group(P<0.05).Conclusion Liangxue Xiaofeng Tang can reduce the gene and protein expression levels of NF-κB pathway protein in PBMC of patients with blood-heat psoriasis and inhibit the activation of NF-κB pathway,which is one of the mechanisms for its treatment of psoriasis.
