摘 要:目的对GB/T 18204.5—2013《公共场所卫生检验方法第5部分:集中空调通风系统》空调冷却水、冷凝水中嗜肺军团菌检验的酸处理方法进行实验条件优化,提高检出效率。方法嗜肺军团菌ATCC33152菌悬液中加入不同浓度及不同体系的酸处理液后反应5 min,再加入不同类型酸反应终止液,最后接种BCYE培养基。采用菌落计数的方法,比较不同处理方式对嗜肺军团菌检出率的影响。结果不同酸浓度下,0.2 mol/L HCl、0.5 mol/L HCl及1 mol/L HCl对嗜肺军团菌检出率影响结果差异无统计学意义(P>0.05)。不同酸处理体系对军团菌检出率组间差异无统计学意义(P>0.05)。不同酸反应终止液对检验结果差异无统计学意义(P>0.05)。结论现有的检验检测方法中,不同的酸浓度、酸处理体系及酸反应终止液对空调冷却水、冷凝水中嗜肺军团菌的影响差异较小。在实际操作中,检验检测人员可选择适合本实验室的方法进行检测。Objective To increase the detection rate of Legionella pneumophila in cooling water and condensate water of air conditioning by optimizing the experimental conditions which described in GB/T 18204.5—2013 Examination Methods for Public Places Part 5:Central air Conditioning Ventilation System.Methods Different concentrations and systems of acid treatment solution were added to Legionella pneumophila ATCC33152 for reaction for 5 min,and then different types of acid reaction termination solution were added,and finally BCYE medium was inoculated.The effects of different treatments on the detection rate of Legionella pneumophila were compared by colony counting method.Results There was no statistical significance on the difference in the effect of 0.2 mol/L HCl,0.5 mol/L HCl and 1 mol/L HCl on the detection rate of Legionella pneumophila.There is no statistical significance on the difference between the different acid treatment systems(P<0.05).There was no statistical significance on the difference in the test results between different acid reaction termination solutions(P>0.05).Conclusion In the existing detection methods,different acid concentrations,different acid treatment systems,and different acid reaction termination liquids have relatively small differences in the effects of Legionella pneumophila in air conditioning condensate and cooling water.In actual operation,the inspector can choose the method suitable for the laboratory.
