摘 要:目的探讨湿润烧伤膏对糖尿病皮肤溃疡小鼠的创面愈合及抗氧化酶中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(gbutathione peroxidase,GSH-px)表达水平的影响。方法自2020年12月至2021年5月,北部战区总医院烧伤整形科选取40只健康小鼠,其中30只小鼠的腹腔内注射链脲佐菌素,诱导成糖尿病小鼠模型,然后将所有小鼠的背部制作溃疡创面。分组:湿润烧伤膏组(实验组)、康复新液对照组(阳性对照组)、模型对照组、空白对照组(健康小鼠),每组10只小鼠。对实验组和阳性对照组的创面分别给予湿润烧伤膏和康复新液进行外用处理,1次/d,连续处理14%d;模型对照组和空白对照组不予处理。2周后,观察小鼠创面的愈合情况及病理变化,采集小鼠球后静脉血,检测血清中抗氧化酶SOD、GSH-PX的表达情况,并比较四组间的差异。结果用药2周后,实验组、阳性对照组创面愈合率明显优于模型对照组,其差异具有统计学意义(P<0.01)。实验组、阳性对照组、模型对照组中SOD、GSH-px的表达与空白对照组比较降低,其差异具有统计学意义(P<0.01),实验组、阳性对照组中SOD、GSH-px的表达与模型对照组比较升高,其差异具有统计学意义(P<0.01)。HE染色结果显示,四组中的成纤维细胞、胶原纤维的再生及排列情况有明显差异。结论湿润烧伤膏处理糖尿病小鼠的溃疡创面后,抗氧化酶SOD、GSH-PX的表达明显升高,具有抗氧化作用。研究表明湿润烧伤膏可促进创面愈合。Objective To investigate the effect of MEBO on healing of skin ulcer in diabetic mice and expression of superoxide dismutase(SOD)and glutathione peroxidase(GSH-px).Methods From December 2020 to May 2021,40 healthy mice were selected.The intraperitoneal injection of streptozotocin was performed in 30 mice,and then the diabetic mouse model was induced.The ulcer was made on the back of all mice.All mice were divided into MEBO group(experimental group),Kangfuxin solution group(positive control group),model control group and blank control group(healthy mice),10 mice in each group.The wounds in experimental group and positive control group were respectively treated by MEBO and Kangfuxin solution for external use,once a day for 14 days.The model control group and blank control group were not treated.After 2 weeks,the wound healing and pathological changes of mice were observed,retrobulbar venous blood was collected,expression of antioxidant enzymes SOD and GSH-Px in serum was detected and the differences among 4 groups were compared.Results At 2 weeks after treatment,the wound healing rate of experimental group and positive control group was significantly better than that of model control group(P<0.01).The expression of SOD and GSH-px in experimental group,positive control group and model control group was lower than that in blank control group(P<0.01).The expression of SOD and GSH-px in experimental group and positive control group was higher than that in model control group(P<0.01).The HE staining showed significant differences in regeneration and arrangement of fibroblasts and collagen fibers among 4 groups.Conclusion After MEBO treatment of ulcer in diabetic mice,the expression of antioxidant enzymes SOD and GSH-px was significantly increased.This study showed that MEBO had antioxidant effect and could promote wound healing.
