摘 要:针对当前GB/T 5889-1986《苎麻化学成分定量分析方法》中用失重法测试脂腊质含量结果不准确、不稳定以及所用提取溶剂苯-乙醇致癌等问题,对分析方法和测试溶剂做出改进。得出如下结论:同样采用苯-乙醇溶剂提取,干重法比失重法测试结果误差小且稳定性好,采用乙醚溶剂替代致癌的苯-乙醇,测试过程更安全、节能。采用单因子和正交试验探索乙醚提取时间、每小时回流次数和试样尺寸对脂蜡质含量测试的影响。测试所得最优方案为:提取时间3 h,回流6~7次/h,试样尺寸1 mm。用红外光谱分析乙醚提取物的成分,其主要成分为脂肪和蜡质,符合脂蜡质含量的测试要求。乙醚回收率在90%左右,回收次数增加对脂蜡质含量的测试结果并无显著影响。In view of the current GB/T 5889-1986 “Quantitative Determination Method for Chemical Constituents in Ramie”, the results of the inaccurate and unstable test results of the tallow wax content by weightloss method, and the carcinogenicity of the extraction solvent benzene-ethanol, etc., the analysis method and testing solvent should be improved. The following conclusions are drawn: the same use of benzene-ethanol solvent extraction, the dry weight method has less error and good stability than the weight loss method, and the use of ether solvent instead of carcinogenic benzene-ethanol makes the test process safer and more energy-saving. Analyze single factor and orthogonal experiments to explore the influence of ether extraction time, reflux times per hour and sample size on the test of lipid wax content. The optimal solution obtained from the test is: the extraction time is 3 h, the reflux is 6~7 times/h, and the sample size is 1 mm. Analyze the composition of the ether extract by infrared spectroscopy. Its main components are fat and wax, which meets the test requirements for fat and wax content. The recovery rate of ether is about 90%, and the increase in the number of recovery has no significant impact on the effect of testing the content of fat and wax.
