摘 要:为了挖掘茎瘤芥响应盐胁迫和根肿菌胁迫过程中发挥功能的生长素响应因子基因(ARF),对进一步研究其在茎瘤芥抗逆过程中的基因功能提供依据。通过生物信息学方法对ARF家族基因启动子顺式作用元件进行分析,采用qPCR的方法对茎瘤芥ARF家族基因在不同器官以及盐胁迫和根肿菌胁迫下的表达模式进行分析。结果表明,在ARF家族基因启动子上发现了多个响应生长素、盐胁迫和病原菌胁迫的顺式作用元件。BjARF1A和BjARF4B在叶中表达水平高,BjARF7C在根中表达水平高。在盐胁迫处理3 h后,BjARF1B和BjARF2B在根中受到强烈诱导表达;处理6 h后,BjARF9A和BjARF13E也受到显著诱导表达。在根肿菌处理条件下,相比于0 h对照,BjARF3A和BjARF3D分别在病原菌处理12,24 h后受到强烈的诱导表达;BjARF13B则在处理后持续下调表达且下调程度逐渐加强。综上所述,筛选到4个显著响应盐胁迫和3个显著响应根肿菌胁迫的ARF家族基因,为进一步研究其基因功能奠定了研究基础。To identify the candidate auxin response factors gene(ARF)responsive to salt and Plasmodiophora brassicae stresses in tuber mustard,and provide a basis for further studying their functions in the resistance of tuber mustard to stresses.Bioinformatic assay was used to analyze the cis-acting elements in the promoters of ARF family genes,and the fluorescent quantitative PCR method was used to analyze the expression patterns of ARF in different organs,as well as their expression levels under salt and Plasmodiophora brassicae stresses.The results showed that several cis-acting elements related to auxin,salt and pathogen stresses were found in the promoters of ARF genes.BjARF1A and BjARF4B were highly expressed in the leaves,and BjARF7C was highly expressed in the roots.After 3 h of salt stress treatment,the expression of BjARF1B and BjARF2B in root were severely induced;after 6 h of salt stress treatment,the expression of BjARF9A and BjARF13E were also significantly induced.Under the pathogen Plasmodiophora brassicae treatment,the expression of BjARF3A and BjARF3D were highly induced by the pathogen at 12,24 h,respectively;while the expression level of BjARF13B were continuously decreased,and the degree of decline gradually increased.In together,four salt stress responsive ARF genes and three Plasmodiophora brassicae responsive ARF genes were found,laying a foundation for the further study of ARF family gene functions.
