摘 要:目的梅毒螺旋体(TP)可引发人体多部位损害和多样病灶,甚至侵入血脑屏障导致神经功能障碍,但其侵袭和致病机制不明。本文采用生物信息学软件分析TP的A型血管性血友病因子基因vwA所编码蛋白的结构和功能,并探讨其在致病过程中可能的作用。方法使用NCBI、CDD、MEROPS、SWISS-MODEL、CDTree、TMHMM、SignalP 4.1等诠释TP的vwA基因,对其编码蛋白进行在线分析。结果TP含有3个A型血管性血友病因子基因vwA1-3,该基因编码蛋白与人血管性血友病因子催化位点相同,均含有GPIbα结合位点和金属离子依赖性黏附位点MIDAS。结论TP含有3个vwA基因,提示TP进入血流后,其合成的vwA1-3基因产物极有可能竞争性地抑制人血管性血友病因子介导的血小板聚集,从而促使其持续性的从破损的血管壁穿越,播散至全身。Objective Treponema pallidum can cause multiple damage and multiple lesions,and even invade the blood-brain barrier and lead to neurological dysfunction.However,its invasion and pathogenesis are still unknown.In this paper,we used bioinformatics software to analyze the structure and function of the protein encoded by vwA of Treponema pallidum,and to explore its potential role in the pathogenesis.Methods NCBI,CDD,MEROPS,SWISS-MODEL,CDTree,TMHMM,SignalP 4.1 were used to interpret the vwA gene of Treponema pallidum and analyze its coding proteins online.Results The whole genome of Treponema pallidum was confirmed to contain three type A von Willebrand factor genes vwA1-3,which encodes the same proteins as the catalytic sites of human von Willebrand factors,and both contain GPIba binding sites and metal ion dependent adhesion sites.Conclusion Treponema pallidum contains three vwA genes,which suggests that when Treponema pallidum enters the bloodstream,the synthesized vwA1-3 gene products are likely to competitively inhibit the platelet aggregation mediated by human von Willebrand factors,so as to promote Treponema pallidum to continuously pass through the damaged blood vessel wall and spread to each organs.
