川芎嗪联合丹参多酚酸对家兔的抗凝血作用及其机制

摘  要:目的探讨川芎嗪与丹参多酚酸不同配比对家兔血液凝固的影响及其作用机制。方法以生理盐水为空白对照组、丹参川芎嗪注射液为阳性对照组,实验组采用川芎嗪与丹参多酚酸注射液,筛选川芎嗪的最佳抗凝农度,与丹参多酚酸配伍后进行联合抗凝血实验,测定体外凝血时间,并在加入兔肺组织浸出液的条件下,观察川芎嗪联合丹参多酚酸对家兔体外凝血时间的影响。结果当川芎嗪终浓度为0.1 mg/mL时,其凝血时间较生理盐水空白对照组明显延长(P<0.01),(395.7±17.2)s VS.(543.7±20.3)s,故川芎嗪在体外抗凝血的最适浓度为0.1 mg/mL;与生理盐水空白对照组比较,丹参川芎嗪阳性对照组的体外凝血时间显著增加(P<0.05)。当川芎嗪(终浓度0.1 mg/mL)与丹参多酚酸(终浓度0.1 mg/mL)的配比为1:2时,实验组的凝血时间较阳性对照组显著延长(P<0.05),(856.0±24.9)s VS.(901.0±7.5)s,为最佳配比;与组织因子对照组比较,加入肺组织浸出液与丹参川芎嗅的阳性对照组的体外凝血时间显著增加(P<0.01)。在川芎嗪与丹参多酚酸1:2配比的基础上加入肺组织浸出液的条件下,其凝血时间较阳性对照组显著延长(P<0.01),(178.3±6.7)s VS.(238.3±3.5)s,实验组对组织因子凝血作用的抑制率为97.4%。结论川芎嗪联合丹参多酚酸可以通过抑制组织因子活性显著提高家兔的抗凝血作用,两者最佳配比为1:2。Objective To investigate the effect and mechanism of different ratio of ligustrazine and salvianolic acids on blood coagulation in rabbits.Methods Normal saline was used as blank control group,and salviae miltiorrhizae and ligustrazine hydrochloride injection was used as positive control group.In the experimental group,the optimal concentration of ligustrazine was selected and combined with salvianolic acids for anticoagulant test using ligustrazine and salvianolic acids for injection.The coagulation rime in vitro was determined.In addirion,the effect of ligustrazine combined with salvianolic acids on the coagulation time in vitro was observed under the condition of adding rabbit lung tissue leach solution.Results When the concentration of ligustrazine was 0.1 mg/mL,the coagulation rime of ligustrazine was significantly longer than that of normal saline blank control group(P<0.01),(395.7±17.2)s VS.(543.7±20.3)s,so the optimal concentration of ligustrazine in vitro andcoagulation was 0.1 mg/mL.Compared with normal saline blank control group,coagulation time in vitro of salviae miltiorrhizae and ligustrazine hydrochloride positive control group was significantly increased(P<0.05).When the ratio of ligustrazine(final concentration 0.1 mg/mL)to salvianolic acids(final concentration 0.1 mg/mL)was 1:2,the coagulation time of the experimental group was significantly longer than that of the positive control group(P<0.05),and the optimal ratio was(856.0±24.9)s VS.(901.0±7.5)s.Compared with the tissue factor control group,the coagulation time in vitro of the positive control group with the addition of lung tissue leach solution and salviae miltiorrhizae and ligustrazine hydrochloride was significandy increased(P<0.01).Compared with the positive control group,the coagulation time was significantly prolonged(P<0.01).When the ratio of ligustrazine to salvianolic acids was 1:2,(178.3±6.7)s VS.(238.3±3.5)s,and the inhibition rate of tissue factor coagulation in the experimental group was 97.4%.Conclusion Ligustrazine comb
