摘  要:目的传统的冷冻保护剂对于精原干细胞(Spermatogonial Stem Cells, SSCs)的保护效果欠佳,该研究在常规冷冻保护剂中添加白藜芦醇(Resveratrol),探究其在SSCs冷冻过程中的作用和机制。方法从小鼠睾丸中分离出精原干细胞,将白藜芦醇添加到冷冻保护剂中,检测冻后细胞存活率;并采用流式细胞技术检测细胞凋亡情况,Western blot检测凋亡蛋白Caspase-3表达情况;并检测冷冻过程中各组活性氧等指标。结果研究发现Resveratrol组细胞存活率显著高于对照组(P<0.05),凋亡率显著低于对照组(P<0.05);Resveratrol组MDA显著低于对照组(P<0.05),T-SOD显著高于对照组(P<0.05);Resveratrol组Caspase-3的表达显著(P<0.05)低于对照组。结论添加Resveratrol的冷冻保护剂可以帮助提高SSCs存活数量,降低细胞凋亡率,这种作用可能与其降低了活性氧水平和Caspase-3的表达有关。Objective Traditional cryoprotectants have poor protective effect on spermatogonial stem cells(SSCs). In this project, resveratrol was added to conventional cryoprotectants to explore its role and mechanism in the freezing process of SSCs.Methods Spermatogonial stem cells were isolated from mouse testis, and resveratrol was added to cryoprotectant;The apoptosis was detected by flow cytometry and the expression of Caspase-3 was detected by Western blot;Active oxygen and other indicators were detected during freezing.Results The results showed that the cell survival rate of resveratrol group was significantly higher than that of the control group(P < 0.05), and the apoptosis rate of resveratrol group was significantly lower than that of the control group(P < 0.05);MDA in resveratrol group was significantly lower than that in control group(P < 0.05), and T-SOD in resveratrol group was significantly higher than that in control group(P < 0.05);The expression of Caspase-3 in resveratrol group was significantly lower than that in control group(P < 0.05).Conclusion The cryoprotectant supplemented with resveratrol can help increase the survival number of SSCs and reduce the apoptosis rate, which may be related to the decrease of reactive oxygen species and Caspase-3 expression.
