摘 要:CONSTANS(CO)基因是植物成花光周期调控路径中重要的调控因子。为探究甜橙(Citrus sinensis)CO基因家族的生物学特性,本研究从甜橙全基因组中筛选到8个CO基因家族成员,依次命名为CsCO1~CsCO8,并对其理化性质和基因表达等进行了分析。结果表明,甜橙CO基因编码蛋白的长度为347~481 aa,分子量在37.93~53.66 kD之间,为亲水蛋白,亚细胞定位于细胞核。系统进化分析发现,CsCOs分为3个亚组,每个亚组的基因结构与Motif相同,所有CsCOs基因序列都含有B-box2和CTT保守结构域。顺式作用元件分析发现,CsCOs启动子序列中含有大量的光响应元件。各组织基因表达结果说明,CsCOs基因在甜橙不同组织中存在表达差异性,各基因之间也存在表达差异。实时荧光定量PCR分析发现,CsCOs基因在两个不同花期甜橙品种花芽的表达趋势相同,呈先升高再下降趋势,检测的CsCO3、CsCO4和CsCO7在早花品种赣南早中多个检测时期升高的量都显著高于同期的正常开花品种‘纽荷尔’。表明CsCOs对甜橙开花时间起重要调控作用。The CONSTANS(CO)gene is an important regulatory factor in the photoperiod regulation of plant flower formation.In order to explore the biological characteristics of the CO gene family of Citrus sinensis,8 CO family members were selected from the genome of Citrus sinensis,named CsCO1~CsCO8 in turn,and their physical and chemical properties and gene expression were analyzed.The results showed that the length of the protein encoded by the CO gene of sweet orange is 347~481 aa,and the molecular weight is between 37.93~53.66 kD.It is an acidic hydrophilic protein and is subcellularly located in the nucleus.Phylogenetic analysis revealed that CsCOs were divided into three subgroups,and the gene structure and motif of each subgroup were the same.All CsCOs gene sequences contained B-box2 and CTT conserved domains.The analysis of cis-acting elements revealed that the CsCOs promoter sequence contains a large number of light-responsive elements.The gene expression results indicated that the expression of CsCOs gene is different in various sweet orange tissues,and there were also differences in the expression of various genes.Real time fluorescence quantitative PCR analysis showed that the expression trend of CsCOs gene in flower buds of two sweet orange varieties at different flowering stages was the same,showing a trend of increasing firstly and then decreasing.But the increased amounts of CsCO3,CsCO4 and CsCO7 in early flowering varieties’Gannanzao’were significantly higher than those in normal flowering varieties’Newhall’at the same stage.The results showed that CSCOs played an important role in regulating the flowering time of sweet oranges.
