摘 要:为了探究橡胶树胶乳中紫色酸性磷酸酶(purple acid phosphatase,PAP)与乙烯刺激增产关联情况。从橡胶树胶乳中鉴定得到9条具有完整保守序列的紫色酸性磷酸酶基因序列,具紫色酸性磷酸酶保守结构域特征。9条序列与木薯、白杨亲缘关系较近,分别属于三类PAP家族,除HbPAP3外均属于高分子量蛋白,蛋白预测定位在多种细胞器中。荧光定量PCR分析组织特异性,结果表明HbPAP3基因在胶乳中表达显著高。在乙烯刺激下,HbPAP26、HbPAP3和HbPAP23基因伴随刺激强度增强在胶乳中基因表达增强3倍以上,推测与乙烯刺激增产和抗氧化活性密切相关。In order to explore the relationship between purple acid phosphatase(PAP)in rubber latex and ethylene stimulation.Nine purple acid phosphatase gene sequences with complete conserved sequences were identified from rubber latex,and they have the characteristics of purple acid phosphatase conserved domains.The nine sequences are closely related to Manihot esculenta and Populus alba,and belong to three types of PAP families respectively.All of them belong to high molecular weight proteins except HbPAP3,and the proteins are predicted to be located in a variety of organelles.Real-time quantitative PCR analysis of tissue specificity showed that the expression of HbPAP3 gene in latex was significantly higher.Under ethylene stimulation,the gene expression of HbPAP26,HbPAP3 and HbPAP23 genes in latex increased by more than three times with increased stimulation intensity,which is speculated to be closely related to the stimulation of ethylene to increase production and antioxidant activity.
