摘  要:G-box结合蛋白(GBF)是一类识别并结合G-box的转录因子,广泛地参与植物基因响应外界刺激的表达调控。本研究采用FPNI-PCR克隆了广藿香醇合酶(PTS)的启动子,并对其序列进行分析,发现其含有G-box等顺式作用元件。为发掘潜在的转录因子,设计引物扩增广藿香中的GBF基因,克隆得到7个GBF基因cDNA序列,并对其进行生物信息学分析。结果表明,克隆得到的GBF均含有bZIP类保守结构域,分别命名为PatbZIP1、PatbZIP2、PatbZIP3、PatbZIP4、PatbZIP5、PatbZIP6、PatbZIP7。通过实时荧光定量PCR技术检测了PatbZIPs在不同组织的表达情况,结果显示出这些GBF在广藿香中的表达量各不相同,有些在不同组织的表达量具有明显区别,表明这些GBF在广藿香生长发育中可能具有不同的功能。这些结果为进一步解析广藿香GBF的功能和作用机理积累了有价值的资料,也为分析GBF与广藿香基因表达调控的关系提供了参考。G-box binding factors(GBF)are a type of transcription factors that recognize and bind to G-box,and widely involved in the expression regulation of plant genes in response to external stimuli.In this study,we analysis the promoter sequence of patchouli synthase gene,and found that contains cis-acting element such as G-box.In order to explore its potential transcription factor,we designed primers to amplify the GBF gene in patchouli,and 7 cDNA sequences of the GBF genes were cloned.Bioinformatics analysis was conducted,and the cloned GBFs contains bZIP conserved domains,named as PatbZIP1,PatbZIP2,PatbZIP3,PatbZIP4,PatbZIP5,PatbZIP6 and PatbZIP7 respectively.The expression of PatbZIPs in different tissues were detected by quantitative real-time PCR.The results showed that the expression of these GBFs in patchouli were different,some of them have significant disparity in different tissues,which indicated that these GBFs may have diverse functions in the growth and development of patchouli.These results have accumulated valuable data for further analysis of the function and mechanism of GBF in patchouli,and provided a reference for the analysis of the relationship between GBF and patchouli gene expression regulation.
