摘  要:为了探究七叶一枝花种子休眠解除过程中差异基因的表达及相关代谢通路,本研究以七叶一枝花不同萌发阶段的种子为材料,通过新一代高通量测序技术对样品进行转录组测序及相关的生物信息学分析。差异基因GO分类分析表明,萌发过程DEGs主要富集在生物过程、细胞成分和分子功能3个方面,包括代谢过程和催化活性等途径。基于KEGG数据分析,20个萌发相关的代谢通路参与种子萌发过程,其中淀粉和糖代谢和植物激素信号转导中差异基因发生显著性变化。在种子萌发阶段,种子休眠被打破是由于β-淀粉酶基因表达发生上调,而休眠解除后,由于α-淀粉酶基因表达上调,促进了种子胚轴发育和胚根发生。此外,KEGG富集分析表明,七叶一枝花种子萌发中八大激素参与该过程,包括生长素、细胞分裂素、赤霉素、脱落酸、油菜素内酯、茉莉酸、水杨酸和乙烯。In order to reveal the expression of differential genes and related metabolic pathways during the dormancy release of Paris polyphylla seeds,the study chose those during different germination stages as materials.The transcriptome sequencing and related bioinformatics analysis were applied through a new generation of high-throughput sequencing technology.The GO classification analysis for differential gene showed that DEGs during seed germination were mainly enriched in three aspects:biological process,cellular component and molecular function,including metabolic process and catalytic activity.Based on the analysis of KEGG data,there are mainly 20 germination related metabolic pathways involved in the process of seed germination.The significant changes of the differential genes happen on starch and glucose metabolism and plant hormone signal transduction.During the germination stage,seed dormancy was broken due to up-regulation ofβ-amylase gene expression,while after dormancy release,the up-regulation ofα-amylase gene expression promoted seed hypocotyl development and radiculogenesis.In addition,the KEGG enrichment analysis revealed that eight hormones were involved in the process of seed germination,including auxin,cytokinin,gibberellin,abscisic acid,brassinolide,jasmonic acid,salicylic acid and ethylene.
