摘  要:沉香是一种名贵中药和天然香料。2-(2-苯乙基)色酮是其主要的特征性成分,聚酮合酶(PKS)是2-(2-苯乙基)色酮类化合物生物合成的关键酶之一。本研究根据转录组数据在白木香中克隆了一个新的聚酮合酶基因,命名为As PKS02。该基因包含一个1185 bp的完整开放阅读框,编码395个氨基酸。As PKS02具有典型的聚酮合酶结构特征,包含1个保守的结构域和7个保守的基序。As PKS02蛋白与马来沉香(Aquilaria malaccensis)中的聚酮合酶蛋白具有91%的同源性,表明与马来沉香具有较近的亲缘关系。AsPKS02基因的编码区包含1个内含子和2个外显子,其启动子区含有12个响应环境胁迫和激素应答的调控元件。进一步的表达分析显示在结香剂处理下,AsPKS02基因的表达于白木香茎干中呈上调趋势,且表达量在第9天高达对照处理的7倍左右。表明As PKS02可能参与了沉香2-(2-苯乙基)色酮的生物合成,为进一步揭示As PKS02在2-(2-苯乙基)色酮积累和沉香形成中的作用提供了参考。Agarwood is a valuable traditional Chinese medicine and natural spice.2-(2-Phenylethyl)chromone is the characteristic constituent of agarwood and polyketide synthase(PKS)is one of the key enzymes in the biosynthesis of 2-(2-phenylethyl)chromone.In this study,a new PKS gene was cloned in Aquilaria sinensis according to the transcriptome data,named as AsPKS02.The gene contains a complete 1185 bp open reading frame encoding395 amino acids.As PKS02 has typical polyketide synthase structure,containing one conserved domain and 7 conserved motifs.The protein homology comparison revealed that As PKS02 protein is in 91%similarity with Aquilaria malaccensis and there is a closer relationship between Aquilaria sinensis and Aquilaria malaccensis.The As PKS02 contained one intron and two exons,and twelve regulatory elements respond to environmental stress and hormone response were identified at the promoter region of AsPKS02.The expression analysis indicated that the expression level of AsPKS02 was up-regulated continuously during inducer reagent treatment from 3 to 9 days in the stem of Aquilaria sinensis plant and its relative expression on the 9 th day was nearly 7 times higher than the control treatment.These results suggest that As PKS02 might be involved in the biosynthesis of 2-(2-phenylethyl)chromone in agarwood,which provide a reference for further revealing its role in the accumulation of 2-(2-phenylethyl)chromone and the formation of agarwood.
