摘  要:目的建立黔产大籽獐牙菜的HPLC指纹图谱分析方法。方法采用高效液相色谱法,MEDJEN Camesil C_(18)柱(5μm,250×4.6 mm)为色谱柱,以25%甲醇的0.1%磷酸溶液-乙腈为流动相进行梯度洗脱,流速为1.0 ml/min,检测波长为254 nm,柱温40℃。结果对11批贵州不同产地的大籽獐牙菜样品进行了分析,标定了共有峰32个,确定了黔产大籽獐牙菜指纹图谱的共有模式,其中4个共有峰得到指认,分别为獐牙菜苦苷、龙胆苦苷、当药苷、芒果苷。结论所建立的方法重复性好,简便准确,可用于黔产大籽獐牙菜的HPLC指纹图谱测定,并可为今后规范其药材资源及质量评价提供科学依据。Objective To establish a HPLC fingerprint analysis method for Swertia macroseed from Guizhou.Methods HPLC analysis was performed on a MEDJEN Camesil C_(18) column(5μm,250×4.6 mm)with the eluting system of gradient consisted of 25%methanol of 0.1%H_(3)PO_(4)-acetonitrile.The flow rate was 1.0 ml/min.The detection wavelength was 254 nm and the column temperature was maintained at 40℃.Results The HPLC fingerprint was established and 11 batches of Swertia macrosperma samples with 32 common peaks were compared.Four peaks were identified as Swertiamain,Gentiopicrin,Sweroside and Mangiferin.Conclusion The method with good reproducibility is simple and accurate,which can be used for determination of HPLC fingerprint and quality control of Swertia macrosperma from Guizhou and the specification for the future medicinal material resources and provide a scientific basis for quality evaluation.
