摘  要:目的:利用生物信息学方法对热带无爪螨第1组分变应原(Blo t 1)特征进行分析。方法:采用SignalP 4.1服务器用于分析Blo t 1的信号肽序列,ProtScale tools对Blo t 1的理化性质进行分析,NetPhos3.1服务器预测分析Blo t 1的磷酸化位点。采用SOPMA服务器预测其二级结构。结果:Blo t 1是一种亲水蛋白,其第1-18位氨基酸为Blo t 1的信号肽序列,磷酸化位点预测显示:Blo t 1含有16个丝氨酸激酶、12个苏氨酸激酶及16个酪氨酸激酶,没有跨膜结构域;Blo t 1的二级结构中包含α螺旋(氨基酸占比28.56%),β转角(氨基酸占比7.95%),延伸链(氨基酸占比19.05%)和无规则卷曲(氨基酸占比44.44%)。Blo t 1的B细胞表位有5个肽序列(38-43,115-118,125-130,156-159,204-212),T细胞表位预测得到3个肽序列(89-109,134-154,247-267)。结论:对Blo t 1的表位分析,为Blo t 1基于表位的疫苗和肽-ELISA的研发奠定理论基础。Objective: Using bioinformatics methods to analyze the characteristics of the Blomia tropicalis group 1 allergen(Blo t 1). Methods: SignalP 4.1 server was used to analyze the signal peptide sequence of Blo t 1. The ProtScale tools was used to analyze the physical and chemical properties of Blo t 1, and the NetPhos3.1 server was used to predict and analyze the phosphorylation sites of Blo t 1. The SOPMA server was used to predict the secondary structure of Blo t 1. Results: Blo t 1 was a hydrophilic protein with stable physical and chemical properties. The amino acids 1-18 were the signal peptide sequence of Blo t 1. The phosphorylation site prediction showed that Blo t 1 contained 16 serine kinases, 12 threonine kinases and 16 tyrosine kinases. Blo t 1 had no transmembrane domain. The secondary structure of Blo t 1 contained α helix(amino acids accounted for 28.56%), β turn(amino acid accounted for7.95%), extension chain(amino acid accounts for 19.05%)and random curl(amino acid accounts for 44.44%). The B cell epitope of Blo t 1 protein had five peptide sequences(38-43,115-118,125-130, 156-159, 204-212), and three peptide sequences(89-109,134-154,247-267). Conclusion: These results provided a theoretical basis for the design of Blo t 1 epitope-based vaccines and peptide-ELISA.
