四妙勇安汤对ApoE-/-小鼠主动脉粥样硬化斑块胶原和血管重构的影响

摘　　要：目的观察四妙勇安汤对载脂蛋白E(ApoE)-/-小鼠主动脉粥样硬化斑块胶原和血管重构的影响。方法将30只ApoE-/-小鼠随机分为模型组、辛伐他汀组和四妙勇安汤组,均以高脂饲料喂养,并将10只C57BL/6J小鼠作为正常对照组。给药19周后,采用天狼星红-苦味酸染色观察组织内胶原纤维类型及分布情况;免疫组织化学染色测定小鼠主动脉根部斑块内兔抗鼠平滑肌细胞α肌动蛋白(SM-αactin)表达量、转化生长因子-β_(1)(TGF-β_(1))及血管内皮生长因子(VEGF)表达。结果与模型组比较,四妙勇安汤可增大斑块内胶原面积,升高TGF-β_(1)表达,但对斑块局部VEGF表达未见明显影响;血管内外弹力膜面积与血管腔面积呈明显缩小趋势,作用强度较辛伐他汀强,但差异无统计学意义。结论四妙勇安汤可抑制斑块内胶原降解,并一定程度上抑制血管病理性重构,发挥稳定动脉粥样硬化斑块的作用。Objective To observe the effects of Simiao Yongan Decoction on collagen and vascular remodeling of aortic atherosclerotic plaque in ApoE-/-mice.Methods Thirty ApoE-/-mice were randomly divided into model group,simvastatin group,and Simiao Yongan Decoction group,all mice were fed high fat diet,and 10 C57BL/6J mice were selected as the normal control group.After 19 weeks,the expressions of smooth muscle cellαactin(sm-αactin),transforming growth factor-β_(1)(TGF-β_(1)),and vascular endothelial growth factor(VEGF)in the aortic root plaques of mice were detected by immunohistochemical staining.Results Compared with the model group,in Simiao Yongan Decoction group,collagen area in the plaque and TGF-β_(1) were increased,without effect on local VEGF expression in the plaque.The elastic membrane area and vascular of lumen area showed a significantly decreasing trend,and the efficacy was better than that of simvastatin,but there was no significant different.Conclusion Simiao Yongan Decoction could inhibit the degradation of collagen and vascular remodeling,to stabilize atherosclerotic plaque.
