摘  要:目的:研究交联聚维酮(PVPP)与黄芩提取物配比之后对黄芩苷含量测定的影响。方法:取不同用量PVPP(0.1、0.2、0.5、0.8、1.0、1.2、1.5、2.0 g)与相同体积黄芩苷溶液进行混合,采用高效液相色谱法测定在不同量交联聚维酮中黄芩苷的含量变化;称取1.5 g PVPP,加入黄芩苷溶液20 mL,分别于5、10、20、40、60、90 min取样检测黄芩苷含量;按照不同制剂配比加入PVPP后,检测黄芩苷的含量;取黄芩苷提取物与PVPP的混合物0.1 g三份,分别加入人工胃液、人工肠液、水等介质进行吸附后,检测黄芩苷含量变化;PVPP与黄芩中黄芩苷静态吸附后加入不同浓度的乙醇进行解析附,检测其吸附与解析情况。结果:黄芩苷与PVPP具有吸附作用,随着PVPP用量的增加,黄芩苷含量随之降低,最高含量下降超80%;随着PVPP在制剂中用量的增加,可检测出的黄芩苷含量呈现逐渐下降,当用量达到20%时,下降比例可达到50%以上。采用乙醇进行解吸附试验,70%乙醇解吸率最高。结论:在制剂的生产过程中应注意PVPP对黄芩苷成分含量的影响。Objective:To study the effect of the ratio of polyvininyl polypyrrodidone(PVPP)and scutellaria baicalensis extract on the determination of baicalin.Methods:Different amounts of PVPP(0.1 g,0.2 g,0.5 g,0.8 g,1.0 g,1.2 g,1.5 g,2.0 g)were mixed with the same volume of baicalin solution.The content of baicalin in different amounts of cross-linked povidone was determined by HPLC;Took 1.5 g PVPP adding to 20 mL of baicalin solution and then determined the content of baicalin after 5 min,10 min,20 min,40 min,60 min and 90 min,respectively.The content of baicalin was detected after adding PVPP according to different preparation ratios;Took 0.1 g mixtures of baicalin extract with PVPP in triplicate,added artificial gastric juice,artificial intestinal juice,water and other media for adsorption,and detected the change of baicalin content;after static adsorption of baicalin in PVPP and scutellaria baicalensis,different concentrations of ethanol were added for analytical adsorption,and the adsorption and analysis were detected.Results:Baicalin had adsorption with PVPP.With the increase of PVPP dosage,the content of baicalin decreased,and the maximum content decreased by more than 80%.With the increase of the dosage of PVPP in the preparation,the detectable baicalin content decreased gradually.When the dosage reached 20%,the decrease proportion could reach more than 50%.The desorption rate of 70%ethanol was the highest.Conclusion:Attention should be paid to the effect of PVPP on the content of baicalin in the production process of the preparation.
