摘  要:建立测定药物中吡拉西坦的吸收光谱法。在酸性溶液中,吡拉西坦与氯膦酸偶氮Ⅲ发生显色反应,生成的新物质在可见光区666 nm和615 nm(正吸收)及538 nm(负吸收)处的吸光度与吡拉西坦的浓度服从朗伯-比尔定律,线性范围为0.03~2.84 mg·L^(-1),检出限为0.026 mg·L^(-1)(538 nm)和0.028 mg·L^(-1)(615 nm、666 nm),表观摩尔吸光系数(κ)为5.87×10^(4)L·mol^(-1)·cm^(-1)(538 nm)、3.34×10^(4)L·mol^(-1)·cm^(-1)(615 nm)和4.10×10^(4)L·mol^(-1)·cm^(-1)(666 nm)。当用双波长法测定时,表观摩尔吸光系数(κ)为9.21×10^(4)L·mol^(-1)·cm^(-1)(538 nm+615 nm)和9.97×10^(4)L·mol^(-1)·cm^(-1)(538 nm+666 nm),检出限均为0.014 mg·L^(-1)。当用三波长法测定时,表观摩尔吸光系数(κ)达1.33×10^(4)L·mol^(-1)·cm^(-1)(538 nm+615 nm+666 nm),检出限为0.009 mg·L^(-1)。以灵敏度最高的三波长法为例,样品的加标回收率为97.2%~103%,相对标准偏差为1.1%~2.8%。Absorption spectrometric method for the determination of piracetam was established.In an acidic solution, the chromogenic reaction of piracetam and clodronate azo Ⅲ occured, the absorbance of the new substance at 666 nm and 615 nm(positive absorption)and 538 nm(negative absorption)wavelengths in the visible region and the concentration of piracetam obeied to Lambert-Beer’s law, their linear range were 0.03~2.84 mg·L^(-1),detection limits were 0.026 mg·L^(-1)(538 nm)and 0.028 mg·L^(-1)(615 nm and 666 nm),and their apparent molar absorption coefficient(κ)were 5.87×10^(4)L·mol^(-1)·cm^(-1)(538 nm),3.34×10^(4)L·mol^(-1)·cm^(-1)(615 nm)and 4.10×10^(4)L·mol^(-1)·cm^(-1)(666 nm).When measured by the dual wavelength absorption spectrometry method, the apparent molar absorption coefficient(κ)were 9.21×10^(4)L·mol^(-1)·cm^(-1)(538 nm+615 nm)and 9.97×10^(4)L·mol^(-1)·cm^(-1)(538 nm+666 nm),the detection limit all were 0.014 mg·L^(-1).When measured by the three-wavelength absorption spectroscopy, the apparent molar absorption coefficient(κ)reached 1.33×10^(4)L·mol^(-1)·cm^(-1)(538 nm+615 nm+666 nm),and the detection limit was 0.009 mg·L^(-1).Take the most sensitive three-wavelength absorption spectrometry as an example, the spiked recovery of the samples was 97.2%to 103%,and the relative standard deviation was 1.1%to 2.8%.The method was simple, rapid and sensitive, and can be used for quality control analysis of piracetam in drugs.
