摘 要:【目的】探讨半仿生提取的黄芪多糖对乙醇诱导大鼠胃黏膜损伤的修复作用及机制。【方法】将50只雄性SD大鼠随机分成正常组,模型组,胃乃安组,黄芪多糖低、高剂量组,每组10只。各组分别灌胃给药,1次/d,连续2 d。末次给药2 h后,除正常组外,其余各组分别按照1.4 m L/只灌胃给予无水乙醇诱导急性胃黏膜损伤。1 h后处死大鼠,取胃进行胃黏膜损伤评分和病理损伤评分,免疫组织化学法检测凋亡信号通路B细胞白血病2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱氨酸天冬氨酸特异性蛋白酶3(Caspase-3)的蛋白表达。【结果】黄芪多糖高、低剂量组均能显著降低胃黏膜的损伤分数和病理损伤评分(P<0.05),呈剂量依赖关系。与正常组比较,模型组大鼠胃黏膜组织中抑凋亡蛋白Bcl-2的表达水平降低(P<0.05),促凋亡蛋白Bax和Caspase-3的表达水平增加(P<0.01)。与模型组比较,黄芪多糖低、高剂量组胃黏膜组织中抑凋亡蛋白Bcl-2的表达水平增加(P<0.05或P<0.01),促凋亡蛋白Bax和Caspase-3的表达水平降低(P<0.05或P<0.01)。【结论】半仿生提取黄芪多糖对乙醇诱导的大鼠胃黏膜损伤具有明显的修复作用,可通过升高抑凋亡蛋白Bcl-2的表达,降低促凋亡蛋白Bax和Caspase-3的表达发挥作用。Objective To explore the protective effect and mechanism of Astragalus polysaccharide(APS)extracted by semi-bionic extraction against ethanol-induced gastric mucosal injury in rats. Methods Fifty male SD rats were randomly divided into the normal group, the model group, the Weinaian group, the APS low-dose group and high-dose group,with 10 rats in each group. Each group was given intragastric administration once a day for 2 consecutive days. Two hours after the last administration,all the groups were given anhydrous ethanol at1.4 mL per mouse to induce acute gastric mucosal injury except for the normal group. One hour later,the rats were executed and the stomachs were taken for scoring of gastric mucosal injury and pathological injury. The protein expression of apoptotic signaling pathway B cell leukemia 2(Bcl-2), Bcl-2 associated X protein(Bax)and cysteine aspartic acid-specific protease 3(Caspase-3)were detected by immunohistochemical method. Results The injury scores and pathological injury scores of gastric mucosa were significantly decreased in both APS lowdose group and high-dose group(P<0.05)in a dose-dependent manner. Compared with the normal group,the expression level of anti-apoptotic protein Bcl-2 in gastric mucosa of the model group was decreased(P<0.05),and the expression levels of pro-apoptotic protein Bax and Caspase-3 were increased(P<0.01). Compared with the model group,the expression level of anti-apoptotic protein Bcl-2 in gastric mucosa tissues of the APS highdose and low-dose groups were increased(P<0.05 or P<0.01), and the expression levels of pro-apoptotic protein Bax and Caspase-3 were decreased(P<0.05 or P<0.01). Conclusion The APS extracted by semi-bionic extraction can significantly repair the ethanol-induced gastric mucosa injury in rats by increasing the expression of anti-apoptotic protein Bcl-2 and decreasing the expression of pro-apoptotic proteins Bax and Caspase-3.
